腺相关病毒载体介导ZsGreen基因通过3种转染途径在心肌组织转染效果的比较

Comparison on three transfection methods for ZsGreen gene expression mediated by adeno-associated virus in the myocardium of rats

摘要:

目的：采用腺相关病毒（Adeno-associated virus，ＡＡＶ）-9为载体，比较３种转染方法对ZsGreen基因在大鼠心肌组织表达的影响。方法：30只雄性SD大鼠随机分为尾静脉（Tail vein，TV）注射组、心肌（Intramyocardium，IM）注射组、冠脉（Intra－coronary，IC）灌注组，采用相应方法在体转染携带报告基因ZsGreen的腺相关病毒-9型（Adeno-associated virus 9-ZsGreen，AAV9-ZsGreen）。2周后荧光显微镜下观察绿色荧光在大鼠ＩＭ组织的表达情况。结果：TV组ＩＭ有少量绿色荧光分布；IM组绿色荧光表达主要局限在ＩＭ注射位点，注射点周边有少量分布；IC组大量绿色荧光在ＩＭ广泛均匀表达。IM及IC组荧光强度均显著高于TV组（P＜0.01），IC组荧光强度高于IM组（P＜0.01）。结论：IC灌注法可使ＡＡＶ介导的外源基因在ＩＭ组织高效、广泛、均匀表达，优于TV注射法及ＩＭ注射法。
