坎地沙坦酯对急性心肌梗死大鼠血管内皮功能的干预影响

Effects of candesartan cilexetil on vascular endothelial function in rats with acute myocardial infarction

摘要:

目的：研究坎地沙坦酯（candesartan cilexetil，CAN）对急性心肌梗死（acute myocardial infarction，AMI）大鼠血管内皮功能的干预。方法：将大鼠随机分为AMI组、ＣＡＮ组和假手术组，干预2周后处死，检测血压（blood pressure，BP）、血一氧化氮（nitric oxide，NO）、血管紧张素Ⅱ（angiotension Ⅱ，AngⅡ）含量、一氧化氮合酶（nitric oxide synthase，NOS）活性、离体胸主动脉条片舒缩功能。结果：与假手术组比较，AMI组血清NO含量、NOS活性降低，胸主动脉内皮依赖性舒张（endothelium-dependent diastole，EDD）功能减退，血浆AngⅡ含量升高，差异有统计学意义（P=0.000 4）。经ＣＡＮ干预后血清NO含量、NOS活性升高达假手术组水平，胸主动脉EDD显著改善（P=0.0009），同时血浆AngⅡ含量升高，各组ＢＰ并无明显变化。结论：ＣＡＮ在不影响ＢＰ的前提下显著改善大鼠AMI后的血管内皮功能。

Abstract:

Objectives：To study the effects of candesartan cilexetil（CAN） on vascular endothelial function in rats with acute myocar－dial infarction. Methods：Rats were randomized into acute myocardial infarction（AMI） group，sham-operation group and CAN group. Rats were sacrificed at two weeks after the intervention and blood pressure（BP），levels of blood nitric oxide（NO） and angiotension Ⅱ（AngⅡ），activities of nitric oxide synthase（NOS），endothelial vasomotor function of isolated thoracic aorta strips were explored. Re－sults：Compared with those in sham-operation group，levels of serum NO and activities of NOS were lower，function of endothelium-dependent diastoles（EDDs） was decreased seriously while levels of plasma AngⅡ were distinctly higher in AMI group，with significant statistical differences（P=0.000 4）. After CAN treatment，levels of serum NO and activities of NOS were obviously higher，almost reaching the similar levels in sham-operation group；meanwhile function of EDDs in isolated aortic strips was improved greatly（P=0.000 9） and levels of plasma AngⅡ were increased significantly，however，BP in all groups was not affected. Conclusions：CAN can greatly improve the disordered endothelial function of rats with acute myocardial infarction without affecting BP.
