培哚普利对异丙肾上腺素诱导的大鼠心肌肥厚中碱性成纤维细胞生长因子-2的影响

Effects of perindopril on bFGF-2 expression in isoproterenol-induced myocardial hypertrophy of rats

摘要:

目的：探讨培哚普利片对异丙肾上腺素（isoproterenol，ISO）所致心肌肥厚大鼠碱性成纤维细胞生长因子-2（basic fi－broblast growth factor，bFGF-2）表达的影响。方法：将研究大鼠分为对照组、模型组及药物干预组，对照组大鼠背部行皮下注射生理盐水；模型组大鼠皮下注射ISO；药物干预组大鼠皮下注射ISO，培哚普利片灌胃，称量计算各组大鼠左心室质量/体质量（left ventricular mass/body mass，LVM/BM）和全心质量/体质量（heart mass/body mass，HM/BM）并进行对比，应用免疫组化及逆转录聚合酶链反应（reverse transcription polymerase chain reaction，RT-PCR）的方法比较各组大鼠左心室中bFGF-2含量、3组大鼠LVM/BM和HM/BM。结果：大鼠LVM/HM、HM/BM比较，对照组<药物干预组<模型组，组间比较差异有统计学意义（Ｆ＝25.220，Ｐ＝0.０００；Ｆ＝76.761，P=0.000）。免疫组化及RT-PCR显示药物干预组中bFGF-2 mRNA 含量高于对照组，但低于模型组（P=0.000，Ｐ＝0.038）。结论：通过皮下注射ISO能成功制备心肌肥厚大鼠模型。bFGF-2对大鼠心肌肥厚具有促进作用，培哚普利可通过降低bFGF-2表达抑制心肌肥厚。

Abstract:

Objective：To explore the effect of perindopril on basic fibroblast growth factor（bFGF-2） expression in isoproterenol（ISO）-induced cardiac hypertrophy of rats. Methods：The research rats were randomly divided into three groups and 10 rats in each group：control group，model group（3mg/（kg·d） isoproterenol for 10 d） and intervention group（3 mg/（kg·d） isoproterenol and 1 mg/（kg·d） perindopril for 10 d）. Left ventricular mass/body mass（LVM/BM） and heart mass/body mass（HM/BM） were detected and compared；in addition，FGF-2 mRNA and protein in left ventricular tissues were analyzed by RT-PCR and immunohistochemistry.
