Objective:To determine the expression and distribution of BACE2 in human pancreas and insulinoma,and to investigate the function of BACE2 in normal pancreas as well as the possible roles of BACE2 in the amyloidogenesis of T2DM. Methods:Human pancreas and insulinoma tissues obtained from the department of general surgery,the first affiliated hospital were used in this study.We have used immunohistochemistry to detect the distribution of BACE2,BACE1 and IAPP in human pancreas and the distribution of BACE2 in human insulinoma. Western blot were applied to examine the protein levels of BACE2 in human pancreas and insulinoma. Results:Immunohistochemical staining showed that BACE2 expressed in human pancreas islets cells as well as insulinoma,BACE1 expressed in pancreas islets. Double stains indicated that BACE2 expressed mainly in islet β-cells,the same as the expression position of IAPP in pancreas. Western blot revealed that both pancreas and insulinoma expressed BACE2,and the protein level in insulinoma was higher. Conclusion:The presence of BACE2 in human pancreas and insulinoma,especially the coexpression of BACE2 and IAPP in islet β-cells suggests that BACE2 may play a potential role in the amyloidogenesis of T2DM.
