Objective: To investigate the expression of special AT rich sequence binding protein 1(SATB1) in hepatocellular carcino-ma and to study the effect of SATB1 on the proliferation,invasion and metastasis of high metastatic hepatic cell line LM3. Methods:Real-time PCR was used to detect the expression of SATB1 in normal hepatic tissue and hepatocellular carcinoma tissue respective-ly. Real-time PCR and Western blot were applied to test the expression of SATB1 in high metastatic hepatic cell line LM3. The expression of SATB1 in high metastatic hepatic cell line LM3 was suppressed by RNA interference technology. MTT,scratch test and Transwell test were performed to study the effect of SATB1 on the proliferation,invasion and metastasis of high metastatic hepatic cell line LM3. Results:The mRNA expression of SATB1 in hepatocellular carcinoma tissue was obviously higher than that in normal hepatic tissue. The mRNA expression of SATB1 was obviously higher in high metastatic hepatic cell line LM3 than in normal hepatic cell line HL-7702. The expression of SATB1 was successfully interfered in LM3. After the interference of SATB1,the abilities of pro-liferating,invading and metastasizing of LM3 cell line were prominently decreased. Conclusion:SATB1 can highly express in hep-atocellular carcinoma and can promote the proliferation,invasion and metastasis of hepatocellular carcinoma cell line.
