    Abstract:

    Objective:To investigate the effect of claudin-1 on the cell adhesion,invasion and metastasis capacity through inhibiting the expression of claudin-1 in SW620 cells. Methods:The expression of claudin-1 in SW620 cell was inhibited through shRNA in-terfering technique to detect its effect on cell adhesion ability. Then Transwell chamber invasion assay and cell scratch test were applied to detect the invasion and metastasis of cells. Results:Low expression of claudin-1 can inhibit adhesion of SW620 cells to laminin,fibronectin,collagen Ⅳand CollagenⅠ,with adhersive inhibition rate of 61%,29%,48%,12% respectively. The cell invasiveness was stronger in SW620-control group than in SW620-shRNA group,with inhibition rate of 70%(P<0.001). After the inhibition of claudin-1,cell migration was significantly reduced. Conclusion:Inhibiting claudin-1 expression could suppress the adhesive and invasive a-bility of SW620 cells.
