    Abstract:

Objective:To study the effects of phosphoinositide 3kinase(PI3K) inhibitor alone and in combination with epidermal growth factor receptor(EGFR) inhibitor on the proliferation and migration of triple-negative breast cancer(TNBC) cell lines. Meth-ods:Effects of PF-04691502(PF),a PI3K inhibitor, alone or in combination with gefitinib,a EGFR inhibitor,on the proliferation and migration of TNBC cell lines(HCC1937 and MDA-MB 231) were measured by the proliferation, colony formation,wound-healing,flow cytometry and ELISA. Results:PF prevented the proliferation, colony formation and migration of TNBC cell lines, inhibited the protein kinase B(PKB or Akt) phosphorylation of these cell lines and retarded the cells in G1 phase(P<0.05). Significant synergistic effects were observed when in combination with gefitinib,a EGFR inhibitor. Conclusions:PI3K inhibitor when used alone could pre-vent the proliferation, colony formation and migration of TNBC cell lines and when used in combination with EGFR inhibitors could form significant synergistic effects, mechanisms of which may relate with it’s inhibition of Akt phosphorylation and G1 phase arrest.
