Objective:To investigate the effect of PJ34,poly(ADP-ribose) polymerase-1(PARP-1) inhibitor,on the biological behavior of human prostate cancer cell line PC-3,including cells proliferation,apoptosis,migration and invasion. Methods:PJ34 with different concentration was used to treat PC-3 cells,CCK-8 assay was used to detect cell proloferation,Western blot was used to detect the expression of PARP-1,annexin V-FITC/IP double-staining method was used to detect cell apoptosis,Transwell method was used to detect changes of cell biological behavior including migration and invasion. Results:After treatment for 72 h,48 h and 24 h,PRAP-1 levels in the PJ34 group were lower than those in the control group(t=4.647,P=0.009 7;t=4.816,P=0.008 5;t=17.28,P<0.000 1). Apoptosis rate in the PJ34 group was (17.64±4.49)%,while in the control group was (3.83±0.74)%,with statistically signifi-cant differences(t=5.261,P=0.006). Differences in migration had statistical significance in two groups(t=23.67,P<0.0001). In addi-tion,cell invasion had statistically significant differences(t=29.31,P<0.000 1). Conclusion:PJ34 can inhibit PC-3 cells' prolifera-tion,apoptosis and invasion by inhibiting expression of PRAP-1.
