Sestrin2(Sesn2),a member of the Sesns family,is a highly conservative stress-induced protein,which can be up-regulated by DNA damage,oxidative stress and hypoxia. The up-regulated Sesn2 acts as an antioxidant and exerts protection effect under a variety of physiological and pathological conditions. This review has described that the regulatory factor p53,nuclear factor-erythroid 2-related factor 2(Nrf2) and hypoxia inducible factor-1(HIF-1) is able to induce the expression of Sesn2,while Sesn2 can regulate downstream AMP-activated protein kinase(AMPK)/mammalian target of rapamycin(mTOR) and autophagy signals,playing an impor-tant role in cardiovascular and metabolic diseases. This review aims to provide a comprehensive understanding of the protective role of Sesn2 in diseases of heart and blood vessels.
