    • Construction of miRNA-451 expression plasmid and its effect on invasion,metastasis and apoptosis in human A549 lung cancer cells

      Abstract:Objective:To explore of the effect of miRNA-451 on invasion,metastasis and apoptosis of human A549 lung cancer cells. Methods:Hsa-miRNA-451 sequence was found in miRBase database and two oligonucleotides were designed according to the se-quence. The targeted gene was annealed and cloned to eukaryotic expression vector plasmid pGenesil-1.1 thus miRNA-451 eukaryotic expression vector was built(pGenesil-1.1-of miRNA-451 plasmid). PGenesil-1.1-miRNA-451 and pGenesil-1.1-control plasmid were transfected into human A549 lung cancer cells and stably expressed pGenesil-1.1-miRNA-451 and pGenesil-1.1-control cell lines were established after being selected by G418. Flow cytometry was employed to detect cell apoptosis,Transwell chamber to detect miRNA-451 cell invasion and cell scratch assay to detect cell metastasis. Results:Sequencing showed that miRNA-451 eukary-otic expression plasmids were successfully constructed. Cell invasion and metastasis were inhibited in stably expressed pGenesil-1.1-miRNA-451 cell lines compared with those in untreated human A549 lung cancer cells and stably expressed pGenesil-1.1-control cell lines,however,there was no change in cell apoptosis(P >0.05). Conclusion:miRNA-451 has significant inhibitory effect on in-vasion and metastasis of human A549 lung cancer cell,however,not on cell apoptosis.

    • Effect of hepatitis B virus X protein mediated by adenovirus on the differentiation of hepatic progenitor cells

      Abstract:Objective:To detect the effect of hepatitis B virus X(HBx) protein on the differentiation of hepatic progenitor cell and to explore whether HBx protein can promote the malignant transformation of hepatic stem cell. Methods:The hepatic progenitor cell Hp14-19 was infected with adenovirus Ad-HBx and Ad-GFP. The expressions of HBx protein in hepatic progenitor cell were con-firmed by RT-PCR and Western blot. The changes of differentiation marker alpha fetal protein(AFP),delta like homolog(DLK),creatine kinase19(CK19),CK18 and albumin(ALB) in the hepatic progenitor cell treated with HBx were evaluated by RT-qPCR and immunofluorescence. The morphological change was observed by PAS staining at 11 d after the inducement. Results:HBx was ex-pressed in the hepatic progenitor cell Hp14-19 infected with Ad-HBx. Compared with those in control group(infected with Ad-GFP),the expressions of early marker DLK,CK19 and AFP were decreased slowly while later marker CK18,ALB were increased slowly in experiment group(infected with Ad-HBx). PAS staining results showed that HBx inhibited glycogen synthesis in HP14.5 cells. Con-clusions:HBx can inhibit the differentiation of hepatic progenitor cell,which maybe involved in the tumorigenesis of HBx-mediated liver cancer.

      Abstract:Objective:To research the mutation of steroidogenic factor-1(SF-1) gene from 46,XY hypospadias including cryptorchidism and to analyze the relationship between genotypes and phenotypes to reveal the action of SF-1 gene in human gonadal differentiation and development. Methods:Sixty 46,XY hypospadias including cryptorchidism subjects without adrenal insufficiency were enrolled as case group and 40 healthy male children with normal genital development were enrolled as normal control group. The SF-1 genes of all subjects were sequenced,then for DNA sequence alignment using standard nuclear BLAST. Results:Heterozygous deletion(c.11835delC) of SF-1 gene was detected in one case. The patient was manifested with female genitalia at birth,perineal hypospa-dias with bilateral cryptorchidism and sever underandrogenization. One single nucleotide polymorphism(SNP) rs1110061(G/C) of SF-1 gene was found. No significant statistical difference in the allele G/C frequency was found between case group and normal control group. Conclusions:SF-1 gene mutation may be one possible cause of 46,XY hypospadias including cryptorchidism,its phenotype is sever testicular dysgensis including sever underandrogenization.

    • Effect of bone marrow mesenchymal stem cells on improving erectile dysfunction in diabetic rats

      Abstract:Objective:To study the effect of bone marrow mesenchymal stem cells(BMMSCs) transplanted into corpus cavernosum on improving erectile dysfunction in diabetic rats. Methods:BMMSCs were separated and cultivated in vitro,then were identified by morphological method and flow cytometry. The diabetic rat model was constructed by intrapertoneal injection of streptozotocin(STZ),then model of diabetes mellitus induced erectile dysfunction(DMED) was sieved by apomorphine(APO). BMMSCs(2×106 pieces) were transplanted into the corpus cavernosum of the rats in DMED model. Two weeks later,the intracorporeal pressure(ICP) and mean arterial blood pressure(MAP) of normal group,diabetic group and treatment group were determined respectively,the surviving and dif-ferentiation of BMMSCs in corpus cavernosum was observed,vessels in corpus cavernosum were counted after staining the tissue by hematoxylin and eosin. Results:The separation and culture of BMMSCs as well as the construction of the rat model with diabetic erectile dysfunction were successful. Many BMMSCs survived in diabetic group under microscope after being transplanted. The results of erectile function test indicated that the ratio of ICP against MAP was higher in treatment group than in diabetic group. There were more vessels in the treatment group[(12.75±1.89) radices/HP] than in diabetic group[(8.05±1.43) radices/HP]. Conclusion:BMMSCs can improve erectile function of diabetic rat.

    • Promotion of apoptosis of CD4+ T and CD8+ T lymphocytes induced by aberrant expression of programmed death ligand-1 in human cervical carcinomas

      Abstract:Objective:To investigate the expression of programmed death ligand-1(PD-L1) in human cervical carcinoma and the association between PD-L1 expression and T cell infiltration and to observe the effect of PD-L1 on apoptosis of active peripheral blood T lymphocytes of patients with cervical cancer in vitro. Methods:PD-L1 and programmed death receptor-1(PD-1) in human cervical carcinoma expressions were determined in 10 cases of normal cervical tissues and 67 cases of cervical carcinoma tissues by immunohistochemistry staining;the apoptosis of tumor infiltrating lymphocytes was examined by TUNEL assay;the number of tumor infil-trating CD4+ T and CD8+ T lymphocytes was determined by immunofluorescent staining and the apoptotic promoting effect of recombi-nant human PD-L1 protein on peripheral blood CD4+ T and CD8+ T lymphocytes was analyzed by flow cytometry. Results:No expression of PD-L1 in normal cervical epithelium was observed but expression of PD-L1 was found in 70%(47/67)cervical cancer tissues. PD-1 expression was found in part of tumor infiltrating lymphocytes. Some tumor infiltrating lymphocytes showed apoptosis. The tumor infiltrating CD8+ T cells were significantly reduced in PD-L1 positive cervical carcinoma cases(P<0.05). In vitro,recombinant hu-man PD-L1 induced apoptosis of activated peripheral blood T lymphocytes of healthy people and cervical cancer patients and the T cell apoptotic rates were significantly higher than those in blank group,which were (32.8±1.4)%,(38.3±1.5)%(P<0.05) and (27.7±1.3)%,(30.9±1.9)%(P<0.05)respectively. Anti-PD-1 antibody depressed PD-L1 induced T cells apoptosis,the apoptotic rates were (29.8±1.6)% and (31.6±1.4)%(P >0.05). PD-L1 induced apoptosis of peripheral blood CD8+ T and CD4+ T cells. Con-clusions:PD-L1 is aberrantly expressed in cervical cancer. The expression of PD-L1 associates with lymphocytic apoptosis and de-crease of tumor infiltrating CD8+ T cells. PD-L1 promotes apoptosis of activated CD4+ T and CD8+ T cells by PD-L1/PD-1 pathway.

    • Effects of 5-azacytidine on methylation state of P16,DAPK and MGMT genes in HL60 cells

      Abstract:Objective:To explore the effect of 5-azacytidine on myeloid leukemia cell lines through detecting methylation state and expression changes of P16,death associated protein kinase(DAPK) and O-6-methylguanine-DNA methyltransferase(MGMT) after 5-azacytidine treatment for acute promyelocytic leukemia cell lines(HL60 cells). Methods:HL60 cells were cultured with 5-azacyti-dine by routine method for 48 h. Then methylation-specific PCR(MSP) and RT-PCR were respectively used to detect gene methyla-tion status and expression changes of P16,DAPKand MGMT before and after being treated with 5-azacytidine. Results:MSP showed hypermethylated P16,DAPK and MGMT genes before 5-azacytidine treatment. Complete demethylation was observed in P16 and DAPK genes and partial demethylation was observed in MGMT gene after HL60 cells being co-cultured with 5-azacytidine. Expression intensities of P16,DAPK and MGMT genes in HL60 cells before and after treatment of 5-azacytidine were (0.22±0.04) vs. (0.64±0.12),(0.36±0.08) vs. (1.27±0.09) and (0.75±0.1) vs. (1.37±0.1) respectively(P<0.05). Conclusions:The CpG islands of P16,DAPK and MGMT genes are hypermethylated in HL60 cells. 5-azacytidine inhibits proliferation of leukemic cells by demethyla-tion of some related genes.

    • Construction of subtracted cDNA library of diferentially expressed genes of multidrug resistant tubercle bacillus by suppression subtracted hybridization

      Abstract:Objective:To build the subtracted cDNA library of differentially expressed genes of multidrug-resistant tubercle bacillus(MDR-TB)and to further discuss the molecular mechanism of MDR-TB. Methods:Tester was MDR-TB and Driver was sensitive tu-berculosis. Suppression subtractive hybridization(SSH) and T/A cloning technology were done to build the subtracted cDNA library of differentially expressed genes of MDR-TB. Results:The subtracted cDNA library of differentially expressed genes of MDR-TB was successfully built and 113 differentially expressed cDNA fragements of MDR-TB were obtained. Sequencing and homology analysis showed that 5 of them were novel cDNA sequences and 5 sequenced genes were reported to be related with MDR in TB. Conclusions:SSH is an effective method for screening new function genes. Many genes both known and unknown are in correlation with MDR in TB. Discovery of these genes provides a solid foundation for the explanation of MDR mechanism in TB.

    • Expression and clinical significance of gene and protein in human renal carcinoma tissues

      Abstract:Objective:To observe the expression and clinical significance of Yes-associated protein(YAP) gene and protein in human renal carcinoma tissues. Methods:The expressions of YAP mRNA were measured by reverse transcription-polymerase chain reac-tion (RT-PCR) and the expressions of YAP protein were detected by immunohistochemistry in 42 cases of human renal carcinoma tis-sues and their corresponding adjacent non-malignant tissues. Results:The positive expression rates of YAP mRNA in renal carcino-ma tissues and adjacent non-malignant tissues were 88.10% and 14.29% respectively. The positive expression rate of YAP protein was higher in renal carcinoma tissues than in adjacent non-malignant tissues(95.23% vs. 30.95%),with statistical differences(P<0.05). Conclusions:The expression of YAP might be associated with the generation and development of renal carcinoma and its abnormal expression can provide references for the diagnosis and treatment of renal carcinoma. Especially,it can establish the theoretical basis for inducement of cell apoptosis and gene therapy of renal carcinoma.

      Abstract:Objective:To set up the rat model of chronic intermittent hypoxia(CIH) under normal pressure,to study the relationship between CIH and gastroesophageal reflux disease(GERD) and to explore the effect of CIH on expression of interleukin-6(IL-6) and high sensitivity-C reactive protein(hs-CRP) in rat. Methods:Twenty-four sex maturation rats were randomly divided into unhandled control group(UC group,normal feeding for 4 weeks),CIH group(CIH for 4 weeks),removal of hypoxia group(RH group,normal feeding for 4 weeks after CIH for 4 weeks). The pH values and the expressions of IL-6 and hs-CRP in the inferior esophageal segment were determined and observed. Results:The inflammation was more obvious in CIH group than in UC group and RH group. The pH values were significantly lowered(P<0.05) and the expressions of IL-6 and hs-CRP were significantly elevated(P<0.01),with no dif-ference between UC group and RH group(P >0.05). Conclusions:CIH may induce the inflammation change of esophagus and IL-6 and hs-CRP participate in GERD’s development.

    • Changes in expressions of serum IL-13,TGF-β1,VEGF and their relationship with airway inflammation and remodeling in patients with asthma

      Abstract:Objective:To discuss whether serum interleukin-13(IL-13),transforming growth factor-β1(TGF-β1),vascular endothe-lial growth factor(VEGF) play a regulatory role in the pathogenesis of chronic air way inflammation and remodeling and to explore its possible way from the perspectives of cytokine. Methods:Totally 36 cases of acute asthma at oneset stage(20 cases of moderate to severe attack and 16 cases of mild attack),36 cases in remission and 36 healthy cases were enrolled. The venous blood was extracted at the elbow. ELISA was used to determine serum IL-13,TGF-β1 and VEGF level;lung function testing was performed on all patients and the results were statistically analyzed. Results:The expressions of serum IL-13,TGF-β1 and VEGF were positively correlated in patients with asthma. At the same time,the higher the serum concentrations of the above three indicators,the lower the forced expiratory volume in the 1st s(FEV1) and the ratio of FEV1 to forced vital capacity(FEV1/FVC%),indicating that asthma becoming more and more serious. The expressions of serum IL-13,TGF-β1 and VEGF were negatively correlated with FEV1 and FEV1/FVC%. Conclu-sions:The network imbalance of IL-13,TGF-β1 and VEGF is one of the main mechanisms for asthma onset. Mutual influence and promotion are found among the three indicators in chronic air way inflammation and remodeling. United detection of serum IL-13,TGF-β1 and VEGF is of great importance in judging disease severity and providing prognosis,auxiliary diagnosis and treatment for patients with asthma.

    • Toxicity of candidate sealants for rupture of membrane in vitro

      Abstract:Objective:To provide safety referrences when sealing preterm premature rupture of the membranes(PPROM) by evaluating the toxicity of 3 tissue adhesives on fetal membranes. Methods:Cyanoacrylate(CA,CA group),fibrin sealant(FS,FS group) and col-lagen(Coll group) were directly laid on pieces of fetal membrane and an untreated piece was chosen as control. After being incubated for 24 h,the patches were fixed,embedded and sectioned for HE staining to observe the structural changes and TUNEL staining was applied to detect the cell apoptosis. The quantity of proteins related with apoptosis(Bax and Bcl-2) was tested by Western blot. Re-sults:(1)HE staining showed that there was consecutive rupture in the amnion layer treated with CA,while the amnion layer was complete in the other three left groups.(2)TUNEL staining showed that there were statistical differences in apoptotic rate between CA group(51.0±1.8)% and Con group(12.1±0.8)%,FS group(16.0±0.8)%,Coll group(13.0±1.0)%,P<0.01. There were statistical dif-ferences in apoptotic rate between FS group and Con group,Coll group,P<0.05. There was no statistical difference in apoptotic rate between Coll group and Con group,P >0.05.(3)The relative grey value of Bax protein was significantly increased in CA group(1.294±0.029) than in FS group(0.421±0.023),Coll group(0.479±0.024) and Con group(0.287±0.017)(P<0.05). The relative grey values of Bcl-2 protein in CA group,FS group,Coll group and Con group were (0.350±0.034),(0.406±0.269),(0.433±0.020),(0.493±0.027) respectively. CA group had lower expression of Bcl-2 than the left groups(P<0.05),also the expression of Bcl-2 was lower in FS group and Coll group than in Con group(P<0.05). No statistical difference in Bcl-2 expression was observed between FS group and Coll group(P >0.05). Conclusions:(1)According to the research,CA is harmful to the amnion layer and not suitable for fe-tal membrane repair.(2)It’s better to choose FS or collagen to repair fetal membrane for safety concern.

    • Effect of TDP on microvessel formation of autogenous fat transplantation in mice

      Abstract:Objective:To investigate the effect of TDP on microvessel formation of autogenous fat transplantation in mice. Methods:Totally 72 female mices were divided into TDP group,insulin group and control group randomly and animal model of fat transplanta-tion was established. The region of autogenous fat transplantation was treated with TDP for 30 mins at a time,3 times pre day in TDP group. Autogenous fat transplantation was treated with insulin(concentration of 300 mU/L) in insulin group,the fat transplantation in control group was untreated.The microvessel density in fat transplantation was determined by immunohistochemical method. Results:Microvessel density was higher in TDP group and insulin group than in control group(P<0.05) at 10 d after the operation. Microves-sel density was higher in TDP group compared with that of control group at 20 d and 28 d after the operation. Conclusion:It shows that TDP could improve survival rate of autogenous fat transplantation by promoting vessel formation.

    • Relationship between levels of β-HCG,HPL and multiple pregnancies in early pregnant women

      Abstract:Objective:To explore the relationship between β-human chorionic gonadotropin(β-HCG),human placental lactogen(HPL) and multiple pregnancies by measuring β-HCG and HPL in early pregnant women. Methods:The serum levels of β-HCG and HPL in 18 women with multiple pregnancies(including two and three fetuses) were measured every two weeks from the 5th week of pregnancy to the 11th week; other 35 women with single fetus in the same period were taken as controls. Results:The serum level of β-HCG was significantly higher in multiple pregnancies group than in single pregnancy group during the 5-9th week of pregnancy (P<0.05). There was no significant difference in serum level of β-HCG between the two groups at the 11th week of pregnancy(P >0.05). The serum level of HPL was not significantly different between the two groups at the 5th week of pregnancy(P >0.05). The serum HPL was significantly higher in multiple pregnancies group than in single pregnancy group(P<0.01) during the 7-11th week of pregnancy. Conclusion:Obvious increase of β-HCG and HPL in early pregnancy indicates multiple pregnancies,which should be alerted.

    • Local and systemic glucocorticoid treatment for sudden hearing loss:a Meta-analysis

      Abstract:Objective:To assess the efficacy and safety of glucocorticoid in treating sudden hearing loss with local and systemic ways. Methods:The data base of PubMed,OVID,CBM,CNKI,VIP,Wanfang were systematically retrieved and the literatures associated with local and systemic glucocorticoid in the treatment of sudden hearing loss between 1971 and 2011 were collected. Literatures were screened according to the pre-established inclusion and exclusion standards and the quality was evaluated strictly by using the Cochrane Handbook 5.0. The data were extracted and analyzed by using RevMan 5.0 Meta-analysis software. Results:Among all the qualified literatures,7 articles were randomized controlled trials. Meta analysis results showed that the odds ratio(OR) of the total ef-fective rate within two groups was 2.01 and 95% confident interval(CI) was between 1.31 and 3.08. Regarding the influence of blood sugar,the statistical results showed that the value of χ2 was 3.592 and the value of P was greater than 0.05. Conlusions:Although there is no significant difference of the adverse effects between local and systemic glucocorticoid treatment,the local glucocorticoid for sudden deafness is more effective than the systemic administration. It can be preferred in clinical treatment.

      Abstract:Objective:To explore the clinical features and treatment of primary empty sella turcica syndrome(PESS). Methods:The clinical data of 33 patients with PESS were analyzed retrospectively and pertinent literatures were reviewed. Results:Some patients (about 42.4%) had no correlative symptoms. The common symptoms were headache,dizziness,hypopsia,visual field defect and crypt-orrhea. Most patients did not need therapy. Conclusions:The clinical symptom of PESS in most patients is not typical and the diag-nosis depends on imaging data. Most patients do not need therapy.

    • Outcome of reoperation for thyroid cancer:clinical analysis of 147 cases

      Abstract:Objective:To analyze the cause of reoperation for differentiated thyroid carcinoma(DTC) and to explore the time and proper surgical patterns for the reoperation. Methods:Protocols of 147 patients undergoing reoperation for thyroid cancer from January 2002 to September 2009 were reviewed retrospectively with the study considering domestic and foreign literatures. The reoperation in-cluded total thyroidectomy for 66 cases,near-total thyroidectomy for 28 cases,subtotal thyroidectomy for 17 cases,unilateral lobecto-my for 1 case and palliactive mass excision for 17 cases. Central lymph node dissection or modified neck dissection for 116 cases were performed at the same time. Results:Pathological results after reoperation confirmed that the rate of residual carcinoma in thyroid tis-sue was 65.6%(82/125) and the rate of residual in lymph node was 62.9%(28/32). The rate of temporarily recurrent laryngeal nerve injury was 0.68%(1/147) and of temporary hypocalcaemia was 6.8%(10/147),without permanent laryngeal recurrent nerve hurt or hypocalcaemia. Conclusions:Inadequate initial operation,local recurrence and cervical lymph node metastasis of thyroid cancer need reoperation sundoubtedly. The optimal treatment is total thyroidectomy with central compartment lymph node dissection or modified neck dissection.

      Abstract:Objective:To comprehend the physical health and the environmental exposure of couples at child bearing age in Chongqing,to investigate the risk factors before pregnancy and to propose measures of eugenics. Methods:Totally 1 510 couples at child bearing age in Chongqing were participated in a questionnaire survey about their general situation,personal and family history of disease. Analysis was made to comprehend the exposure of risk factors before pregnancy. Results:According to the questionnaire results,32.91% of the female at child bearing age were more than 35 years old;the percentages of the family genetic history in male and female were 76.31% and 34.30% respectively;the highest risk factor for couples at child bearing age was work,41.13% in male and 19.87% in female. Conclusions:The female more than 35 years old should pay more attention to prenatal care to prevent the adverse pregnancy outcomes. The couples who had family genetic history need to focus on health and eugenic counseling. Government need to strengthen the publicity of prenatal and postnatal care to arouse the awareness of the prenatal and postnatal care for couples at child bearing age and to urge them to participate in counseling and pre-pregnancy examination to reduce the birth defects and other adverse pregnancy outcomes.

      Abstract:Objective:To understand the prevalence of depression and anxiety disorder in patients with cardiovascular disease in hos-pitals at primary level. Methods:Totally 510 patients with cardiovascular disease in outpatient and inpatient department were enrolled and investigated with a multicenter cross-sectional design. Using hospital anxiety and depression scale(HADS) to screen the pos-itive patients,who were then assessed with the Hamilton depression and anxiety disorder scale. Results:The results of Hamilton showed that the corrected prevalences for depression disorder,anxiety disorder and depression and anxiety disorder were 19.10%,13.59% and 10.92%. The recognition rate of the treatment of depression and anxiety was 4%. Conclusions:Cardiovascular disease with depression and anxiety disorder is a common problem in hospitals at primary level,its correct identification and treatment is not optimistic.

      Abstract:Objective:To investigate the infection status of hepatitis B virus and the immune state of children admitted to our hospital. Methods:Totally 21 473 children in the outpatient department,hospitalized department and the physical examination center of the children’s hospital of Chongqing medical university in 2009 were enrolled in the study. Statistical analysis on the examination results of HBV serology markers was conducted in clinical laboratory center. Results:The positive rate of HBsAg was 1.4%(301/21 473) in children,including 1.1%(228/21 473) with both positive HBsAg and positive HBeAg and 0.3%(73/21 473) with positive HBsAg but negative HBeAg. The positive rate of anti-HBs was 51.2%(10 990/21 473). The positive rate of anti-HBe and/or anti-HBc was 7.0%(1 501/21 473). The negative rate of HBV serologic markers was 40.4%(8 681/21 473). Conclusions:In 2009,the HBV infection rate of children in Chongqing was 1.4%,lower than that of common crowds in the whole country in 2005. The positive rate of anti-HBs was 51.2%,higher than that of common crowds in the whole country in 2005. The positive rate of HBsAg and/or HBeAg,especially anti-HBs is relatively high,frequent follow-up should be conducted in children.

    • Survey and analysis of adult outpatients’dental anxiety in hospital of stomatology

      Abstract:Objective:To evaluate the incidence and prevalence of dental anxiety in adult outpatients and to discuss the related in-ducing factors. Methods:A questionaire survey was conducted with 300 adult outpatients,aged older than 18. All participants com-pleted the modified dental anxiety scale(MDAS). Level of dental anxiety was evaluated by MDAS. Results:The mean MDAS score was (11.98±2.34). Prevalence rate of dental anxiety was 36.4%. Significant statistical differences of the incidence of dental anxiety were found in regarding to sex,marital status,economic income,education level,self-rated oral health status,being initial treatment or not(P<0.05);but not to nationality and age. Logistic regression analysis demonstrated that the risk factors of dental anxiety were sex,age,economic income and self-rated oral health status. Conclusions:The incidence of dental anxiety is high for the oral adult out-patients. The incidence of dental anxiety is positively correlated with sex,age,marital status,economic income,education level,self-rated oral health status,being initial treatment or not.

    • Study on the status of wedge-shaped defects among urban adults in Ningxia

      Abstract:Objective:To investigate the status of wedge-shaped defects among adults in Ningxia,thus to provide clinical references for the prevention of this disease. Methods:Totally 4 064 adults in Ningxia were investigated in this study according to the criterion issued by China oral health epidemiology survey protocol. The incidence and therapy rate of wedge-shaped defects in different groups were calculated. SPSS 11.5 statistical software was applied to analyze the results. Results:①The overall incidence of wedge-shaped defects was 42.67%. The incidences were increased with the growth of age and there were significant differences among different age groups. It was peaked at the age between 50 and 59,but was decreased at the age between 60 and 69 and more than 70. The over-all therapy rate was 15.63%. The therapy rates were reduced with the growth of age and the differences were statistically significant among different age groups. ②The incidence was higher in male than in female without statistical differences. The therapy rate was high in male than in female with statistical differences. ③The place with the highest incidence of wedge-shaped defects was the first premolar(15.59%),while that with the lowest incidence was the second-molar(0.26%). The differences in the different teeth were statistically significant. The incidence of wedge-shaped defects was higher in maxilla teeth than in mandible teeth and the incidence was higher in the left teeth than in the right teeth with significant differences. The therapy rate of the first premolar was the highest,and the differences in the different teeth were statistically significant.④The method of teeth-brushing was significantly related with wedge-shaped defects. Conclusions:The incidence of wedge-shaped defect is related with tooth location and age in Ningxia. Its incidence is higher than the therapy rate. So it is necessary to strengthen the propaganda of oral cavity of prevention and protection for adults in urban of Ningxia.

    • Characteristics of postoperative life quality change in patients with lung cancer and it’s influencing factors

      Abstract:Objectives:To research into the characteristics of changes of life quality and symptom at 4 months postoperatively in pa-tients with lung cancer and to analyze its influencing factors. Methods:The life quality of 113 patients with lung cancer at the 1st, 2nd and 4th months after surgery was assessed by using the Chinese version of European organization for research and treatment of cancer quality of life questionnaire-core 30(EORTC QLQ-C30) and EORTC QLQ-lung cancer 13(EORTC QLQ-LC13) scale and the data were analyzed by adopting descriptive statistics and multiple linear regression. Results:At the 4 th months,the function and overall life quality of patients were improved significantly except cognitive function(P<0.05). Fatigue,dyspnoea,appetite loss,cough-ing,chest pain,pain in the arm or shoulder were alleviated significantly(P<0.05),however,stabling pain in the hands and feet was worsened(P<0.05),other symptoms has no significant changes. The influencing factors of life quality included depression,smoking history and adjuvant therapy. Conclusions:At the 4th month after the surgery,the overall life quality is improved and most symptoms are eased,however,fatigue and role function obstacle are still existed and should be attached importance to.
