    • Mechanism of osteogenic differentiation for mesenchymal stem cells induced by BMP9 and Wnt3a in vitro

      Abstract:Objective:To analyze the molecular mechanism of osteogenic differentiation of mesenchymal stem cells(MSCs) induced by canonical Wnt3a signal pathway and bone morphogenetic protein 9(BMP9). Methods:C3H10T1/2 cells were infected by recombinant adenovirus expressing BMP9 and Wnt3a,and then alkaline phosphatase(ALP),the early osteogenic marker,was detected by quantita-tive assay. Osteogenic differentiation marker at later stage,calcium deposition was determined by Alizarin Red S staining. The protein levels of osteocalcin(OC) and osteopontin(OPN) were detected by immunocytochemical staining. The expression levels of OC,OPN and Runx2 were analyzed by real time PCR. Results:Wnt3a enhanced the ability of BMP9-induced ALP activity(F=264.962,P=0.000;t(BMP9+Wnt3a)-BMP9=7.19,P=0.000;t(BMP9+Wnt3a)-Wnt3a=16.36,P=0.000) and increased expressions of OC(F=3 920.102,P=0.000;t(BMP9+Wnt3a)-BMP9=39.10,P=0.000;t(BMP9+Wnt3a)-Wnt3a=62.56,P=0.000) and OPN(F=1 935.824,P=0.000;t(BMP9+Wnt3a)-BMP9=40.88,P=0.000;t(BMP9+Wnt3a)-Wnt3a=56.42,P=0.000),with increase of mineral calcium deposition in vitro. Furthermore,Wnt3a increased the mRNA level of Runx2(F=3 635.980,P=0.000;t(BMP9+Wnt3a)-BMP9=79.37,P=0.000;t(BMP9+Wnt3a)-Wnt3a=86.96,P=0.000). Conclusion:Wnt3a,possibly through Runx2,acts synergistically on BMP9 induced osteogenic differentiation of MSCs.

    • Inhibitive effect of dominant negative human epidermal growth factor receptor on the invasion and metastasis of gastric cancer cells

      Abstract:Objective:To explore the effect of dominant negative epidermal growth factor receptor(DNEGFR) on the invasion and metastasis of human gastric cancer cells and to elucidate its molecular mechanism. Methods:The pEGFPN1-DNEGFR was construct-ed and human gastric cancer cells were transfected stably. The invasion and metastasis in vitro were tested by Transwell invasion cham-ber assay. Matrix metalloproteinase-2(MMP-2) and Matrix metalloproteinase-9(MMP-9) mRNA levels of cells were detected by re-versed transcription-polymerase chain reaction(RT-PCR). MMP-2 and MMP-9 protein levels of culture were detected by enzyme-linked immunosorbent assay(ELISA). Results:pEGFPN1-DNEGFR was constructed successfully,human gastric cancer cells was transfected stably. MMP-2 and MMP-9 mRNA levels of cells were down-regulated;MMP-2 and MMP-9 protein levels of culture were down-regulated as well. Conclusions:DNEGFR represses the invasion and metastasis of human gastric cancer cells by inhibiting them from secreting MMP-2 and MMP-9,which lays a solid foundation for further research of DNEGFR in gastric cancer biotherapy.

    • Correlation between single nucleotide polymorphisms at three sites of Klotho gene with coronary heart disease

      Abstract:Objective:To investigate single nucleotide polymorphisms(SNPS) at three sites of the Klotho(KL) gene and to analyze their correlation with coronary heart disease(CHD). Methods:Totally 317 patients were randomly selected as CHD group and 301 healthy people as normal controls. Genomic DNA of leukocytes was extracted from venous blood and the single allele specific primer PCR technique(SASP-PCR) was used to detect allelic genotypes at three SNPS sites of KL gene in order to analyze their relationship with CHD. Results:The frequency of allele AA genotype of KL G-395A in CHD group was obviously higher than that in normal con-trol group(P<0.05). According to the results of sex stratification analysis,the frequency of allele AA genotype in the CHD male sub-group was higher than that in normal control group(P<0.05). The frequency of allele CC genotype of KL C370S was higher than that in normal control group(P<0.05),but the frequency of allele CS genotype was lower than that in normal control group(P<0.05). Accord-ing to the results of sex stratification analysis,the frequency of allele CC genotype in CHD male subgroup was higher than that in normal control group(P<0.05),but the frequency of allele CS genotype was lower than that in normal control male subgroup(P<0.05). There was no significant difference in polymorphic FF,FV and VV frequency in the F-352V gene between CHD group and normal control group(P >0.05). According to the results of combination genotype analysis,the allele genotype frequency of KL G-395A+F352V+C370C,KL A-395A+F352V+C370C and KL A-395A+F352V+C370S in CHD group was higher than that in normal control group(P<0.05),but the allele genotype frequency of KL G-395A+F352V+C370S was lower than that in normal control group(P<0.05). According to the results of sex stratification analysis,the allele genotype frequency of KL G-395A+F352V+C370C,KL A395-A+F352V+C370C was higher than that in normal control male subgroup(P<0.05),but the allele genotype frequency of KL G-395A+F352V+C370S was lower than that in normal control male subgroup(P<0.05). The allele genotype frequency of KL A-395A+F352V+C370S in CHD female subgroup was obviously higher than that in normal control female subgroup(P<0.05). Conclu-sions:This study shows that the male patients who carried with allele genotype KLG-395A SNP AA have higher risk in suffering from CHD. The male patients with allele CS genotype of KL C370S have lower risk in suffering from CHD. This study also finds that the male patients who carried with KL G395-A+F352V+C370C,KL A-395A+F352V+C370C and the female patients who carried with KL A-395A+F352V+C370S have higher risk in suffering from CHD,however,the male patients with KL G-395A+F352V+C370S have lower risk in suffering from CHD.

    • Improvement of hyperoxia-induced pulmonary fibrosis by bone marrow mesenchymal stem cells

      Abstract:Objective:To investigate the effects of bone marrow mesenchymal stem cells(BM-MSCs) on the pulmonary fibrosis in neonatal rats suffering from hyperoxia. Methods:BM-MSCs were isolated from male SD rats and transplanted into receptor rats. The receptor rats were devided into normal control group,normal BM-MSCs group,hyperoxia control group and hyperoxia BM-MSCs group. On the 14th day,the rats were killed and pulmonary tissue samples were taken for pathological examination and hydroxyproline level test. DNA was isolated from female pulmonary tissues and Sry gene expression was detected by reverse-transcription polymerase chain reaction(RT-PCR). Results:Pathological examination showed significant fibrosis and structural disorder in pulmonary tissue of hyperoxia control group. The hydroxyproline level in pulmonary tissue samples was significantly higher in hyperoxia control group than in normal control group. The thickness of alveolar wall and the content of hydroxyproline in the lung were reduced significantly in hyperoxia BM-MSCs group. The results from PCR indicated that Sry gene was detected in normal BM-MSCs group and hyperoxia BM-MSCs group,however,the Sry gene expression was increased in hyperoxia BM-MSCs group compared with that in normal BM-MSCs group. Conclusion:BM-MSCs could ameliorate pulmonary fibrosis in neonatal rats suffering from hyperoxia.

    • Construction of recombinant adenoviral vector encoding human LMP-1 and its expression in osteosarcoma cell U2OS

      Abstract:Objective:To construct recombinant adenoviral vector encoding human LIM mineralization protein-1(LMP-1) and to in-vestigate its expression in osteosarcoma cell line U2OS. Methods:The cDNA of LMP-1 was amplified by PCR from K562 cell line and cloned into pGEM-T,then subcloned into shuttle plasmid pAdtrack-CMV. After restrictive enzyme digestion and DNA sequencing,the recombinant shuttle plasmid pAdtrack-LMP-1 was transferred into HEK-293T cells,which were detected by fluorescent microscopy,RT-qPCR and Western blot to confirm the expression of LMP-1. Then linearized pAdtrack-LMP-1 was homologously recombined with pAdEasy-1 in BJ5183 competent cell to transform into Ad-LMP-1 which subsequently transfected HEK-293A cells under the mediation of lipofectamine. Ad-LMP-1 was packaged and amplified in HEK-293A cells. After Ad-LMP-1 infection,the gene and protein expressions of LMP-1 in U2OS were identified by fluorescent microscopy,RT-qPCR and Western blot. Results:Restrictive enzyme digestion and DNA sequencing analysis revealed that the shuttle plasmid pAdtrack-LMP-1 was constructed successfully. Fluorescence microscopy found that expressed green fluorescent protein was distributed in cytoplasm of transfected HEK-293T. RT-qPCR and Western blot verified LMP-1 to be highly expressed in HEK-293T. After purification,the virus titer reached 1.5×109 pfu/ml. The result of fluorescent microscopy,RT-qPCR and Western blot indicated that LMP-1 mRNA and protein expression were remark-ably increased in U2OS infected with Ad-LMP-1. Conclusion:The recombinant adenovirus Ad-LMP-1 has been successfully con-structed and can be helpful for the further research involved in LMP-1.

    • Effects of Artemisinin extract on the cardiovascular and respiratory function in rabbits

      Abstract:Objective:To observe the effects of Artemisinin extract on the cardiovascular and respiratory function in rabbits. Methods:Thirty-two healthy New Zealand white rabbits were randomly assigned to four groups. 10 ml/kg normal saline,10,100 mg/kg and 500 mg/kg Artemisinin extract were injected respectively into the rats in the four groups. Arterial blood pressure,respiratory movement,electrocardiogram,blood gas analysis and histological examination of the lungs in the experimental rabbits were measured on 12th d after the intravenous injection. Results:Compared with those in the control group,(1)there was no difference in blood pressure,res-piratory movement,heart rate and electrocardiogram among each experimental group(P >0.05);(2)the values of pH,PaCO2,PaO2,HCO3ˉ,BE and SaO2 were not statistically changed in low and middle dosage groups(P >0.05),but the PaO2 value was decreased sig-nificantly in high dosage group(P<0.05). Conclusions:Artemisinin extract will not exert obvious adverse effect on the cardiovas-cular system in rabbits,but the pulmonary inflammatory response can be induced by high-dose Artemisinin extract in experimental rabbits.

    • Effects of Ketamine on lipopolysaccharide-induced nuclear factor-kappaB expression of human umbilical vein endothelial cells

      Abstract:Objective:To investigate the effects of Ketamine on lipopolysaccharide(LPS)-induced translocation of nuclear factor-kappaB(NF-κB) in human umbilical vein endothelial cell nuclei and to explore it’s anti-inflammatory action. Methods:Human um-bilical vein endothelial cells(HUVECs) were divided into four groups:group C(blank control group),group L(1 μg/ml LPS was added),group KL1(3 μg/ml ketamine and 1 μg/ml LPS were added) and group KL2(30 μg/ml ketamine and 1 μg/ml LPS were added). The tanslocation and expressions of NF-κB p65 in endothelial cell nuclei were detected by Western blot,reverse transcription polymerase chain reaction(RT-PCR) and image analysis system of HPIAS 2000. Results:The tanslocation and expression of NF-κB p65 in en-dothelial cell nuclei induced by LPS were reduced in group KL1 and KL2. Conclusions:The tanslocation and expression of NF-κB p65 in endothelial cell nuclei can be induced by LPS and Ketamine can suppress the activation.There are positive correlations be-tween Ketamine concentration and its inhibitory effect. The anti-inflammatory effect of Ketamine may be achieved by the translocation of ?资B.

    • Preliminary study on the regulatory effect of hepatitis B virus on Pasha promoter activity

      Abstract:Objective:To investigate the regulatory effect of hepatitis B virus (HBV) on the expression of Pahsa and to discuss its mechanism. Methods:RT-PCR and Western blot were performed to check Pasha expression in HepG2 and HepG2-H7 cell lines. Pasha promoter luciferase reporter plasmid,pGL3-Pasha-P,was constructed and transiently transfected into HepG2 and HepG2-H7 cells to anlyze the activity of Pasha promoter. Then HepG2 cells were transiently cotransfected with 0.5 μg of pGL3-Pasha-P and HBx,HBs,HBp,HBc expression plasmids,respectively and the luciferase activity was detected. Results:The results of RT-PCR and Western blot showed that the expressions of Pasha in HepG2-H7 cells were lower than those in HepG2 cells. The luciferase activity assay of Pasha promoter demonstrated that HBV could suppress Pasha promoter activity and HBx and HBs proteins may play impor-tant roles in this process. Conclusion:HBV can down-regulate Pasha expression in HepG2-H7 cells through inhibiting its promoter activity.

    • Expression of SATB1 in hepatocellular carcinoma and its promoting effect on hepatocellular carcinoma cell proliferation and metastasis

      Abstract:Objective: To investigate the expression of special AT rich sequence binding protein 1(SATB1) in hepatocellular carcino-ma and to study the effect of SATB1 on the proliferation,invasion and metastasis of high metastatic hepatic cell line LM3. Methods:Real-time PCR was used to detect the expression of SATB1 in normal hepatic tissue and hepatocellular carcinoma tissue respective-ly. Real-time PCR and Western blot were applied to test the expression of SATB1 in high metastatic hepatic cell line LM3. The expression of SATB1 in high metastatic hepatic cell line LM3 was suppressed by RNA interference technology. MTT,scratch test and Transwell test were performed to study the effect of SATB1 on the proliferation,invasion and metastasis of high metastatic hepatic cell line LM3. Results:The mRNA expression of SATB1 in hepatocellular carcinoma tissue was obviously higher than that in normal hepatic tissue. The mRNA expression of SATB1 was obviously higher in high metastatic hepatic cell line LM3 than in normal hepatic cell line HL-7702. The expression of SATB1 was successfully interfered in LM3. After the interference of SATB1,the abilities of pro-liferating,invading and metastasizing of LM3 cell line were prominently decreased. Conclusion:SATB1 can highly express in hep-atocellular carcinoma and can promote the proliferation,invasion and metastasis of hepatocellular carcinoma cell line.

    • Research on the relative risk factors for genomic DNA methylation in Wistar rats

      Abstract:To discuss the effect of hyperhomocysteinemia(HHCY) and hypercholesterolemia(HTC) on genomic DNA methylation and DNA methyltransferase activity in aortic tissue of Wistar rats and to compare their differences. Methods:Totally 33 Wistar rats were equally randomized into three groups:control group,methionine group and high-cholesterol group(n=11). The rats in control group were fed with normal chaw while those in the other two groups were fed with formula chaw for three months. Heart blood was drawn to detect the serum homocysteine(Hcy) and total cholesterol(TC). Aortic genomic DNA was extracted to detect genomic DNA methylation levels and aortic nucleoprotein was extracted to detect DNA methyltransferase activity. Results:The results of Dunnett-t test showed that serum TC level was significantly higher in high-cholesterol group than in control and methionine groups(P<0.05),but there was no statistically significant difference in triglyceride(TG),low density lipoprotein cholesterol(LDL-C) and high density lipoprotein cholesterol(HDL-C) levels between high-cholesterol group and the other two groups(P >0.05). Serum Hcy was significantly higher in methionine group than in high-cholesterol and control groups(P<0.05). Compared with control group,HHCY and HTC significantly increased DNA methyltransferase activity and promoted DNA demethylation in both high-cholesterol and methionine groups(P<0.05),though there was no significant difference between high-cholesterol and methionine groups(P >0.05). Conclusions:Hypomethylation induced by HTC is one of the important mechanisms for the development of atherosclerosis. There is no significant difference between HHCY and HTC in leading hypomethylation.

    • Expression and significance of HMGB1 and E-cadherin in ovarian carcinoma

      Abstract:Objective:To explore the expression and clinical significance of high mobility group box-1(HMGB1) and E-cadherin in ovarian carcinoma. Methods:Between May 2008 and January 2011,immunohistochemical SP method was used to detect the expres-sion of HMGB1 and E-cadherin in 75 patients with ovarian carcinoma and 40 normal ovarian tissues and to analyze the clinical pathological significance of HMGB1 and E-cadherin in ovarian carcinoma. Results:The positive expression of E-cadherin was lower in ovarian carcinoma than in normal ovarian tissues(χ2=12.62,P<0.01). The positive expression of HMGB1 was higher in ovarian car-cinoma than in normal ovarian tissues(58/75 vs. 4/40, χ2=47.60,P<0.01). The expression pattern of HMGB1 and E-cadherin were associated with clinical stage and lymph node metastasis(P<0.05),but not with histological subtypes and histological stages(P >0.05). The expression of HMGB1 was negatively correlated with E-cadherin(r=-0.299,P<0.01) in ovarian cancer. Conclusion:HMGB1 and E-cadherin might play an important role in the carcinogenesis and metastasis of ovarian carcinoma.

    • Experiment study of correlation between CT scan parameters and noise of different tissues

      Abstract:Objective:To investigate the correlations between CT scan parameters and noises of different tissues and to discuss the factors influencing the image quality. Methods:The anthropomorphic phantom equivalent to human tissue was scanned by 16 slices spiral scanner(GE BrightSpeed) with different voltages(80,100,120 kV and 140 kV),tube currents(80,100,120,180,240 mA and 300 mA) and pitches(0.562,0.938,1.375 and 1.75). 120 kV,300 mA and 0.938(pitch) were using as the standard. One of the three scanning parameters was changed and other scanning parameters were fixed. The CT value and image noise were recorded and an-alyzed(Standard derivation,SD). Results:There was no statistical difference in noises of the lung among groups of 80,100,120,140 kV(F=0.966,P >0.05). There were statistical differences between groups of 80 kV and 120 kV in noises of the chest wall,soft tissue of spine and aorta,P<0.05. The noises of different tissues were statistically different among groups of 80,100,120,180,240 mA and 300 mA(Lung F=3.28,P<0.05;soft tissue of spine F=11.89,P<0.05;aorta F=196.67,P<0.05). There were statistical differences be-tween 80 mA group and 300 mA group in noises of the lung,chest wall,soft tissue of spine and aorta,P<0.05. The noises of the lung,soft tissues of chest wall and spine were not statistically different among groups with different pitches(0.562,0.938,1.35 and 1.75). (Lung F=0.23,P >0.05;chest wall F=1.53,P >0.05;soft tissue of spine F=2.27,P >0.05). There were statistical differences in noises of aorta among groups with different pitches,F=9.68,P<0.05. Conclusions:The noises of different tissues are increasing with the de-crease of tube voltage,tube current and increase of pitch,but the increase of noise in the lung is not obviously. The chest scan with low dose can reduce the radiation dose and keep the image quality without obviously increasing the noise in the lung.

    • Effects of Simvastatin on glial fibrillary acidic protein expression in rats with traumatic brain injury

      Abstract:Objective:To study the changing rules of glial fibrillary acidic protein(GFAP) expressions in the rat brain after traumatic brain injury(TBI) and to observe the effect of Simvastatin(SIM)on them based on the TBI rat model. Methods:Totally 54 Sprague-Dwalye(SD) rats aged 5 weeks were employed and divided into sham TBI group,control group and treatment group(n=18). TBI model was established in control group and treatment group by Feeney’s method. The rats in treatment group were fed SIM 10 mg/kg in the evening pre-injury and in every evening post-injury while those in control group were fed the same dose of starch each time. Then the rats were sacrificed and the rat brains were collected at different time points(3,12,24 h and 3,7,14 d post the injury). The changes of GFAP expressions in cornu ammonis(CA) area were detected with immunohistochemistry. Results:The GFAP positive cell percentage was higher in control group than in sham TBI group at 3 h after the injury. The GFAP positive cell percentage reached the peak at 3 d and 7 d after the injury(P<0.01). The positive expression of GFAP was significantly lowered in treatment group than in control group at 7 d after the treatment(P<0.05). There was no difference at 1,2,3 d and 14 d after the treatment between con-trol group and treatment group(P >0.05). Conclusion:SIM treatment could decrease the over-expression of GFAP in rats after TBI.

    • Minimally invasive percutaneous plate osteosynthesis combined with locking compression plate in the treatment of distal tibial fracture

      Abstract:Objective:To observe clinical efficacy of minimally invasive percutaneous plate osteosynthesis(MIPPO) combined with locking compression plate(LCP) in the treatment of distal tibial fracture. Methods:Totally 20 patients with distal tibial fracture were treated by MIPPO combined with LCP from January 2009 to February 2011. The fracture healing,functional recovery and complication incidences were evaluated by clinical examination and radiographs. Results:All cases were followed up for 10-22 months(averaged 13 months). All patients had bony union with an average recovery time of 16 weeks. No case of screw loose or fixation failure was observed;2 cases(10%) suffered from angles malunion(less than 7°);6 cases(30%) suffered from claudication postoperatively;4 cases(20%) experienced the reduce of the motion range of the injured ankle by at least 20° than the other one;3 cases(15%) can not return to work;1 cases(5%) had infection at later stage,but cured after the treatment. According to Baird-Jackson scoring system,13 cases were graded as excellent,4 cases as good,2 cases as fair,1 case as poor and the excellent and good rate was 85%. Conclusion:With the advantages of minimally invasiveness,solid fixation,quick recovery and fewercomplications,MIPPO combined with LCP is effective in the treatment of distal tibial fracture thus need promoting in clinics.

    • Comparative analysis and bias assessment on results of partial biochemical items detected by different biochemical testing systems

      Abstract:Objective:To investigate the comparability among different biochemical testing systems in the same laboratory by perform-ing the comparative analysis and bias estimation on the results. Methods:According to NCCLS document EP9-A2,3 biochemical test-ing systems including system 2 and 3(Y1-Y2,comparative testing systems) and system 1(X,targeted testing system,which was com-posed of Hitachi 7600 biochemical analyzer P1,maker reagents,c.f.a.s. calibrator and BIO RAD quality control) were used to obtain correlation coefficient and linear regression equation. The values of aspartate aminotransferase(AST),alamine aminotransferase(ALT),gamma-glutamyl transferase(GGT),alkaline phosphatase(ALP),Urea,creatinine(Cr) and uric acid(UA) in patient’s serum were de-tected by the three systems. Then the coefficient and the equation were used to evaluate the system bias between comparative testing systems and targeted testing system. The comparability of different testing systems was judged by half of the CLIA’88 standard. Re-sults:The results of the three testing systems were significantly correlated(r2>0.95). The results of all testing items showed good comparability except those of Urea(with low concentration level) tested by system 2(Y2) and system 1(X) (%SE>1/2CILA’88 TEa). Conclusions:There is good comparability among the results of AST、ALT、GGT、ALP、Cr、UA detected by different testing systems in our hospital except those of Urea with low concentration level.

Methods:Totally 120 patients with uterine leiomyomas were randomly divided into three groups. Oxytocin at concentrations of 0.12,0.24 U/min and 0.36 U/min were infused respectively into patients in the three groups. Fibroid feeding arteries changes before and after the intravenous in-fusion of Oxytocin were observed by using color doppler ultrasound. The differences of arterial changes responding to the use of Oxytocin in all three groups were analyzed. Indicators of blood pressure,pulse,respiration and adverse events were recorded dur-ing the procedure and within 3 months of follow-up. Results:Peak systolic velocity,mean blood flow velocity,diameter and blood flow of the fibroid feeding arteries of patients in all three group were decreased,but resistance index was increased with statistical differ-ences compared with that before the infusion(P<0.05). But there was no significant statistical difference of arterial changes before and after the intravenous infusion of Oxytocin among the three groups(P >0.05). No adverse event was observed in all patients. Conclu-sions:Oxytocin can reduce blood supply of the fibroids. 0.12 U/min Oxytocin intravenous infusion could be the most favorable dosage for ultrasound ablation in the treatment of uterine fibroids.

      Abstract:Objective:To investigate the efficacy and safety of applying Telbivudine in the treatment of chronic hepatitis B(CHB) for consecutive 3 years. Methods:Totally 62 patients with CHB who received monotherapy of Telbivudine in the department of infectious diseases in the second affiliated hospital of Chongqing medical university took Telbivudine 600 mg orally once per day. The virology,serology and biochemical indicators of 62 patients were detected and analyzed before and at the 156th week after the treatment. The 62 patients who received the treatment for 3 years were divided into two groups according to the hepatitis B virus(HBV) DNA level(<lg3 copy/ml and ≥lg3 copy/ml) and comparison was made at the 156th week during the treatment in order to know the ef-ficacy and safety of the treatment. Chi-square test or calibration chi-square test was applied to do the efficacy comparison. Results:After 156 weeks of treatment,HBV DNA negative rate was 79.0%,Alanine aminotransferase(ALT) normalization rate was 72.6%,HBeAg seroconversion rate was 39.7% and drug resistant rate was 11.3%.       Abstract:Objective:To assess the prognosis after loop electrosurgical excision procedure(LEEP) of cervical intraepithelial neopla-siaⅡ-Ⅲ(CINⅡ-Ⅲ) with positive margins by human papillomavirus(HPV) genotyping in order to guide clinical treatment. Meth-ods:Totally 65 cases (9.26%) were found positive margins in 702 cases of CINⅡ-Ⅲ after LEEP. ThinPrep cell test(TCT),colpo-scope and HPV genotype were performed preoperatively and at every 6 months after LEEP. Results:The recurrence rate of positive margins was higher than that of negative margins(P<0.05). The recurrence was not correlated with HPV subtype,multiple HPV types and lesions grade in incision margins(P >0.05). But the persistence HPV infection was associated with higher recurrence rates at 6 months after the operation(P<0.05). Conclusion:HPV genotype may help to detect recurrences in CINⅡ-Ⅲ with positive margins after LEEP.

    • Comparative study of spasmolytic cocktail in preventing radial artery spasm during percutaneous coronary intervention through radial artery

      Abstract:Objective:To compare the efficacy of spasmolytic cocktail composed of Nitroglycerin,Verapamil,Diltiazem and Isosorbide mononitrate in reducing the incidence of radial atery spasm in patients undergoing transradial coronary interventions. Methods:Pa-tients enrolled were randomly divided into three groups. After successful cannulation and sheath insertion into radial arteries,patients in group A received intra-aterial Nitroglycerin 200 μg and Verapamil 200 μg,patients in group B received Isosorbide mononitrate 5 mg and Verapamil 200 μg,patients in group C received Nitroglycerin 200 μg and Diltiazem 2.5 mg. Then the incidence of spasm was determined. Results:There were significant statistical differences between groups A and C(P<0.05). Conclusion:Intra-arterial pre-medication with cocktail composed of 200 μg Nitroglycerin and 200 μg Verapamil is more effective in preventing radial spasms dur-ing percutaneous coronary interventions through radial artery.

    • Observation on the clinical efficacy of glue in the treatment of pulmonary bullae under CT guide

      Abstract:Objective:To evaluate the efficacy and safety of OB glue in the treatment of pulmonary bullae under CT guide and to seek for the new treating methods. Methods:Sixty patients with pulmonary bullae were enrolled and received preoperative examination. glue was injected into the pulmonary bullae under CT guide. and six minutes walk test were regularly applied after the surgery. The intraoperative and postoperative complications,the changes of thoracic CT and the improvement of pulmonary function were observed. Results:The intraoperative and postoperative complications included chest pain(96.7%),pneumothorax(25%),hemoptysis(21.7%) and fever(21.7%). The results of thoracic CT showed that the curative rate was 96.7% and the obvious effect rate was 3.4%. Six-minute walk test was performed in 30 cases and the pulmonary function was improved. Conclusion:Percutaneous pulmonary puncture and OB glue in the treatment of pulmonary bullae under CT guide is a secure and efficient therapy.

    • Observation on the efficacy of Sanyrene combined with Iodophors for diaper dermatitis in elderly patients

      Abstract:Objective:To explore a new preventing and treating method for diaper dermatitis in elderly patients. Methods:Totally 68 elderly patients with diaper dermatitis were included in the present study. Diaper dermatitis was caused by fecal and urine incontinence resulted from various factors. The patients were randomly divided into two groups(n=34). The buttocks of all patients were thor-oughly cleaned with warm water and soft towels or wet wipes at the beginning of the study. In Sanyrene and Iodophors group(obser-vation group),0.5% Iodophor was evenly rubbed into the flushed skin outside-in,then was cleaned with normal saline after being dried,then Sanyrene was evenly sprayed into the flushed skin after being dired. In Sanyrene group(control group),Sanyrene was well sprayed into the flushed skin according to the methods above. The procedure was performed respectively in both groups three times a day and the efficacy and healing time were observed. Results:Significant differences were found in the curative rate and healing time between the two groups(P<0.05). Conclusion:Sanyrene combined with Iodophor is an effective method for preventing and treating diaper dermatitis in elderly patients.
