    • Role of P38 mitogen-activated protein kinases signaling pathway in the differentiation of bone morphogenetic protein 9 induced C3H10T1/2 stem cell into myocardiocytes

      Abstract:Objective:To investigate the role of P38 mitogen-activated protein kinases(P38MAPK) signal pathway in the differentiation of bone morphogenetic protein 9(BMP9) induced C3H10T1/2 stem cell into myocardiocytes. Methods:The phospho-P38MAPK and P38MAPK were measured by Western blot at 24 h after the C3H10T1/2 stem cell being transfected with pAdEasyBMP9 and the phos-pho-P38MAPK was detected by immunofluorescence at 24 h after the C3H10T1/2 stem cell being transfected with pAdEasyBMP9. The cardiac-specific proteins cTnT and CX43 were measured by Western blot,the cardiac-specific proteins cTnT and α-MHC were measured by immunofluorescence,the cardiac-specific gene GATA4 and MEF2C expressions were detected by RT-qPCR at 21 d after the C3H10T1/2 stem cell being transfected with pAdEasyBMP9 and treated with SB203580. Results:Transfection with pAdEasyBMP9 produced rapid increase in phospho-P38MAPK and little change in P38MAPK. Phospho-P38MAPK expressed in nucleus and cytoplasm or in different states of nucleus. SB203580 significantly inhibited the pAdEasyBMP9-induced expression of CX43,cTnT,α-MHC,GATA4,MEF2C in C3H10T1/2 stem cell. Conclusions:P38MAPK signal pathway can be activated in C3H10T1/2 stem cell after being transfected with pAdEasyBMP9 and can promote the myocardiocytes differentiation.

    • Effects of Berberine on lipid metabolism and atherosclerosis in hyperlipidemic rabbits and its mechanism

      Abstract:Objective:To investigate the effects of Berberine(Ber) on lipids metabolism and atherosclerosis in hyperlipidemic rabbits and to study the possible mechanism. Methods:The hyperlipidemic rabbit model was established by feeding high fat diet for 2 months. 40 New-Zealand rabbits were randomly divided into control group,high fat diet model group,high fat diet and Ber treated group[28,112 mg/(kg·d)],high fat diet and Fenofibrate treated group[30 mg/(kg·d)]. The levels of serum total cholesterol(TC),triglycerides(TG),low density lipoprotein-cholesterol(LDL-C) and high density lipoprotein-cholesterol(HDL-C) were determined after 2 months,pathomorphological changes of aortic arteries was observed and inductible nitric oxide synthase(iNOS) and scavenger receptor class-B type 1(SR-B1) mRNA expression in aortic tissue were quantified by quantitative real time PCR. Results:The levels of serum TC,TG and LDL-C were obviously elevated(P<0.05),apparent atherosclerotic plaque was observed in aortic arteries and iNOS and SR-B1 mRNA expressions were obviously increased(P<0.05) in high fat diet group compared with those in control group. Serum TC,TG and LDL-C levels were decreased(P<0.05),HDL-C level was increased(P<0.05),degree of aortic atherosclerosis was mitigated,foam cells were reduced and iNOS activity and SR-B1 mRNA expressions were reduced(P<0.05) in high fat diet and Ber treated group than in high fat diet model group. Conclusions:Ber can obviously reduce the blood lipid levels and aortic atherosclerosis in the hyperlipidemic rabbits. The mechanisms may be related with the down-regulation of the iNOS and SR-B1 mRNA expressions.

    • Effect of stiffness of substrate on vascular endothelial cell growth and proliferation

      Abstract:Objective:To clarify the effect of substrate stiffness on the growth and biological activity of human umbilical vein endothe-lial cells(HUVECs) through studying the mechanical properties of substrate for cell growth and inoculating HUVECs to substrates with different stiffness. Methods:Substrates with different stiffness were built by using bisacrylamide and acrylamide compounds. The sub-strates were sterilized and were cross-linked by collagen to inoculate HUVECs. MTT assay,phalloidin cell staining and flow cytometry analysis were performed to detect the cell cycle. Results:With the enhancement of base stiffness,the proliferation of HUVECs was inhibited significantly(P<0.01) and the cell morphology and growth cycle were also changed accordingly. Conclusions:The changes in substrate stiffness have great impact on the growth and proliferation of HUVECs,which gives further biomechanical evidence to ex-plain the atherosclerotic vascular endothelial cell morphology and dysfunction.

    • Construction and expression of recombinant adenovirus vector carrying transforming growth factor-β3 gene

      Abstract:Objective:To construct the recombinant adenovirus vector containing transforming growth factor-β3(TGF-β3) and en-hanced green fluorescent protein(EGFP) in order to transfect rabbit bone marrow mesenchymal stem cells(MSCs). Methods:The cD-NA TGF-β3 was integrated into the shuttle vector of pAdxsi-CMV with EGFP,then the product was transfected into package cell HEK 293 and the recombinant adenovirus vector expressing TGF-β3 gene was generated. The expression of EGFP was identified by PCR and fluorescent microscopy. The titer of pAdxsi-EGFP-TGF-β3 was tested with end-paint dilution assay and its tranfection ef-ficacy was examed. The expression of TGF-β3 was detected by Western blot. Results:The recombinant adenovirus vector pAdxsi-EGFP-TGF-β3 containing TGF-β3 and EGFP was constructed successfully. PCR analysis showed that TGF-β3 cDNA was inserted into the recombinant adenoviral plasmid. The recombinant virus vector was collected by packaging HEK293 cells. The titer of pAdxsi-EGFP-TGF-β3 was 2×1010 pfu/ml. Western blot demonstrated that TGF-β3 was expressed in rabbit bone marrow MSCs. Conclusions:EGFP-TGF-β3 recombinant adenovirus vector is successful constructed and TGF-β3 protein is effectively expressed in rabbit bone marrow MSCs,which provides the basis for research of gene therapy in preventing hypertrophic scar after wound healing.

    • Effects of surgery on expressions of serum anti-survivin antibody and significance of combined detection of anti-survivin,anti-p53 and anti-c-myc antibody in laryngeal squamous cell carcinoma

      Abstract:Objective:To exam the serum anti-survivin antibody levels in post-surgery and pre-surgery samples of patients with laryngeal squamous cell carcinoma(LSCC) and to investigate the potential use of anti-survivin,anti-p53,and anti-c-myc antibodies in the monitoring of LSCC postoperatively. Methods:Enzyme-linked immunosorbent assay(ELISA) was used for the detection of serum anti-survivin,anti-p53,and anti-c-myc antibodies. Results:(1)After radical surgical treatment,the anti-survivin,anti-p53,and anti-c-myc antibodies expression levels were decreased significantly. (2)High levels of anti-survivin antibody were detected in the serum sample of patients with LSCC,especially in samples from individuals with cervical lymphatic metastasis.(3)High positive detection rates were obtained through independent testing of anti-survivin,anti-p53 and anti-c-myc antibodies;the combined positive rate of all three antibodies was inferior to that of anti-survivin antibody but superior to that of anti-p53 and anti-c-myc antibodies. Conclusions:This indicates that anti-survivin antibody may be a biomarker of LSCC. Combined testing of three different tumor markers may be valuable for monitoring of the surgery efficacy.

    • Experimental study on preventive effects of testosterone on knee osteoarthritis in male rabbits

      Abstract:Objective:To investigate the preventive effect of testosterone on knee osteoarthritis in male rabbits. Methods:Eighteen male rabbits were randomly divided into castration group,noncastration group and testosterone supply group(n=6). The rabbits in cas-tration group underwent the excision of anterior cruciate ligment and the medial meniscus on the right knee joint while those in non-castration group and testosterone supply group underwent the excision of bilateral testicles along with the treatment in castration group. Testosterone undecanoate(6 mg/kg,once every two weeks,three times in total) was injected intramuscularly to rabbits in testosterone supply group at the third week after the operation. All rabbits were killed by gas embolism at the eighth week. Macroscopic evaluation was performed on the articular cartilage of medial malleolus of femur. Insulin-like growth factors(IGF-1) immunohistochemical examination was performed in order to investigate the expression of IGF-1 in articular cartilage and the im-proved Mankin’s scores was applied to test the serum levels of total testosterone at different stages. Results:Expressions of IGF-1 were higher in noncastration group and testosterone supply group than in castration group(P<0.05). Mankin’s scores were lower in noncastration group and testosterone supply group than in castration group(P<0.05). The serum level of testosterone was lower in cas-tration group than in noncastration group and testosterone supply group at the fourth and eighth week(P<0.05). Conclusion:Testos-terone has preventive effect on knee osteoarthritis in male rabbits.

    • Effects of prostaglandin E2 on matrix metalloproteinase-2 activity in synovial cells and its signaling pathway

      Abstract:Objective:To investigate the effects of prostaglandin E2(PGE2) on matrix metalloproteinase-2(MMP-2) activity in syn-ovial cells and its signaling pathway so as to help discuss the possible effect of PGE2 on the healing ability of anterior cruciate liga-ment(ACL) after injury. Methods:Cultured synovial cells were treated with different concentrations of PGE2,and gelatin zymography assay was performed to detect MMP-2 activity at 12,24,48 h and 72 h after treatment.In presence of 10 ng/ml PGE2,each of seven signal pathway-specific inhibitors was respectively used to treat the synovial cells and MMP-2 activity was detected and semi-quan-tified. Results:PGE2 significantly elevated MMP-2 activity in synovial cells in a dose and time dependent manner. Inhibition of the p38 pathway with SB203850,ERK pathway with PD98059,and PKA pathway with KT5720 had no specific effect on PGE2-induced MMP-2 activity. However,inhibition of the JNK pathway with SP600125,AP-1 pathway with Curcumin,and nuclear factor-κB(NF-κB) pathway with Bay11-7082 and-7085 dramatically decreased PGE2-induced MMP-2 activity to 43%,25%,16% and 11%,respec-tively(P<0.01). Conclusions:PGE2-induced MMP-2 activity in synovial cells may account for ACL’s poor healing ability after in-jury and this process might be regulated via JNK,AP-1 and NF-κB pathway. Inhibition of NF-κB pathway has the strongest decreas-ing effect on PGE2-induced MMP-2 activity,which provides a novel strategy for ACL repair.

    • Effect of Budesonide on transform growth factor-β1 induced vascular endothelial growth factor release by lung fibroblasts

      Abstract:Objective:To observe the effect of glucocorticoid(Budesonide) on TGF-β1 induced human fetal lung fibroblasts-1(HFL-1) secretion and vascular endothelial growth factor(VEGF) pro-duction and to discuss its mechanism. Methods:HFL-1 were cultured in vitro and were treated with various concentrations of Budesonide in the presence of TGF-β1 stimulation. Post-culture media were collected and VEGF content was tested by enzyme-linked immunosorbent(ELISA). The cell lysates were subjected to Western blot analysis to test the protein expressions of phosphor-ERK MAPK and phosphor-Smad3. Results:Budesonide can induce VEGF release by HFL-1 in a dose-dependent manner(P<0.05). Budesonide with high concentration can suppress the TGF-β1 induced VEGF release(P<0.05). The protein expressions of phosphor-ERK MAPK and phosphor-Smad3 were decreased after Budesonide pre-treatment(P<0.05). Conclusions:Budesonide may reduce TGF-β1 induced VEGF production in the lung,probably through the Smad/ERK signaling pathway,which may provide new sight into the molecular mechanism underlying glucocorticoids therapy for airway inflammatory diseases.

    • Evaluation of hepatocellular carcinoma by combined detection of serum AFP-IgM complex and liver function

      Abstract:Objective:To analyze the value of the combined detection of a new hepatocellular carcinoma(HCC) marker,alpha-feto-protein immunoglobulin M(AFP-IgM) complex and liver function in diagnosing HCC. Methods:The serum from 64 cases of HCC and 27 cases of liver cirrhosis was collected. ELISA and electrochemiluminescence were employed in the detection of AFP-IgM complex and AFP respectively. The clinical data of patients’ liver function were collected. Results:According to the receiver operating char-acteristic(ROC) curve,the best cut-off value of AFP-IgM immune complex was 300 AU/ml. At this best cut-off value,the serum AFP-IgM complex was significantly higher in the patients with tumor diameter ≤3 cm than in the patients with tumor diameter >3 cm((1 195.4±561.8) AU/ml vs. (584.3±771.7) AU/ml). The serum AFP-IgM complex in patients at Ⅰ-Ⅱ clinic stages was higher than that in patients at Ⅲ-Ⅳ clinic stages((1 033.0±508.0) AU/ml vs. (569.2±791.7) AU/ml)(P<0.01). According to ROC curve,the best cut-off value of gamma-glutamyl transpeptidase(GGT) was ≥154 U/L. The sensitivity of GGT in the diagnosis of HCC was 73.4%,with specificity of 55.6%,and the sensitivity of combined AFP-IgM and GGT in the diagnosis of HCC was 46.9%,with specificity of primary 70.3% and effective rate of 53.8%. Conclusions:AFP-IgM detection helps to improve the effectiveness in the diagnosis of small hepatocellular carcinoma and HCC at early stage. AFP-IgM combined liver function tests can help to do differential diagnosis of liver cirrhosis and HCC,which is expected to become screening method for small hepatocellular carcino-ma at early stage.

    • Isolation,cultivation and differentiation of rabbit bone marrow mesenchymal stem cells in vitro

      Abstract:Objective:To investigate the isolation and cultivation of rabbit bone marrow mesenchymal stem cells(rBMSCs) in vitro,to observe their biological characteristics and to discuss the paths of inducing the rBMSCs into osteoblasts and lipoblasts in order to pro-vide experimental basis for selecting seed cells. Methods:The rabbit was put to death by air embolism to the ear marginal vein with the anesthesia of urethane. The femur of the rabbits was got under the sterilized condition. rBMSCs were separated by percoll den-sity gradient centrifugation method and the whole bone marrow adherence culture method in vitro. The morphology of cells was ob-served by inverted microscope. At the third passage,rBMSCs were chosen to be induced into osteoblasts and lipoblasts. After 14 d of incubation,alkaline phosphatase detection was used in osteoblast-induced group and oil red O staining was performed in lipoblast-induced group. Results:The rBMSCs with good reproductive activities were obtained successfully by percoll density gradient lentrifu-gation and the whole bone marrow adherence culture method. The biological characteristics of the cells in the third generation were i-dentical. In osteoblast-induced group,alkaline phosphatase test showed strong positive expression. After oil red O staining,there was a large number of intracytoplasmic lipid droplets stained red in lipoblast-induced group. Conclusions:rBMSCs can be isolat-ed and proliferated effectively by percoll density gradient cen-trifugation method and the whole bone marrow adherence culture method.The cultured rBMSCs grow up and reproduce stably and could be induced and differentiated into osteoblasts and lipoblasts. So rBMSCs are excellent seed cells in tissue engineering.

    • Expression and clinical significance of RRM1 protein in non-small cell lung cancer at stage of Ⅱb/Ⅲa

      Abstract:Objective:To investigate the postoperative expression of ribonucleotide reductase subunit M1(RRM1) protein in non-small cell lung cancer(NSCLC) at stage of Ⅱ b/Ⅲ and to discuss the effect of RRM1 protein expression on disease free survival(DFS) of patients underwent adjuvant chemotherapy. Methods:Totally 100 patients with NSCLC at stage of Ⅱb/Ⅲa who underwent radical surgery were included in this study. The expression of RRM1 protein in tumor specimens was assayed by streptavidin-perosidase(SP)immunohistochemistry retrospectively. Correlations between the expression of RRM1 protein and the clinical features or DFS time af-ter operation were analyzed. SPSS 13.0 was used to analyze the effect of RRM1 protein expression on the patients’ prognosis. Results:(1)No difference in the expression of RRM1 protein was found among patients with different genders,ages,smoking habits,patho-logical differentiation and stages(P>0.05). RRM1 protein expression was increased in squamous carcinoma than in non-squamous car-cinoma although there was no significant difference(44.1% vs. 30.3%,P=0.172);(2)Patients with lower RRM1 protein expression ex-perienced longer DFS time compared with those with higher RRM1 protein expression(median DFS,14 months vs. 8 months,P=0.019 4);(3)In patients with adjuvant chemotherapy of Gemcitabine/Cisplatin,squamous carcinoma led to longer DFS time than non squamous carcinoma(median DFS,14 months vs. 11 months,P=0.011 9);lower RRM1 protein expression led to longer DFS time than higher RRM1 protein expression(median DFS,13 months vs. 6 months,P=0.021 7). No difference in the length of DFS time was found in the patients with only surgery(median DFS,15.5 months vs. 13 months,P >0.05). (4)COX multivariate regression analysis showed that the expression of RRM1 protein and pathological type were the independent prognostic factors of DFS in NSCLC at stage of Ⅱb/Ⅲa. Conclusion:RRM1 protein is a valuable predictive factor for Gemcitabine/Cisplatin adjuvant chemotherapy in NSCLC at stage of Ⅱb/Ⅲa.

    • Killing activity of antigen-pulsed dendritic cells combined with cytokine induced killer cells against hepatocarcinoma cells

      Abstract:Objective:To observe the proliferation activities,phenotype changes and killing activities of liver cell antigen-pulsed den-dritic cells(Ag-DC) cultured with cytokine-induced killer(CIK) cells against HepG2 hepatocarcinoma cells. Methods:Peripheral blood mononuclear cells(PBMNC) were isolated from healthy donors,then DC and CIK cells were induced. The Ag-DC was co-cul-tured with CIK(Ag-DC-CIK) and the phenotype and proliferation of CIK and Ag-DC-CIK cells were observed. Their cytotoxicity ac-tivities against HepG2 hepatocarcinoma cells were detected by MTT assay. Results:The proliferation of Ag-DC-CIK cell group was significantly enhanced and high expressions of double positive cells of CD3+CD8+ and CD3+CD56+ were observed. The proliferation and cytotoxic activity were higher in Ag-DC-CIK cells than in CIK cells(P<0.05). Conclusion:The proliferation and liver cancer cell-killing activity of Ag-DC-CIK cells which obtained from co-culturing of Ag-DC and CIK cells are significantly higher compared with those of CIK cells.

    • Correlation between Galectin-3 and Wnt pathway-related protein in salivary gland tumor

      Abstract:Objective:To study the expression of Galectin-3,Wnt pathway related protein(β-catenin,Cyclin D1 and E-cadherin) in salivary gland tumors and to explore the relationship and the clinical significance among them. Methods:The benign and malignant tumors of salivary gland specimens were observed under HE staining and light microscopy. Immunohistochemical staining(S-P method) was applied to detect the expression and correlation of Galectin-3,β-catenin,E-cadherin and Cyclin D1 in 107 cases of salivary gland tumors(58 cases of malignant tumors and 49 cases of benign tumors) and 38 cases of chronic inflammation of salivary glands were enndled as negative control. Results:The positive rate of Galectin-3 was 72.4%(42/58) in malignant salivary tumor,higher than that in benign salivary tumor(P<0.05). The positive rates of β-catenin and Cyclin D1 protein were higher in malignant salivary tumor[75.9%(44/58) and 81%(47/58)] than in benign salivary tumor(P<0.05). There were significant positive correlations be-tween Galectin-3 and β-catenin(r=0.313,P<0.05). Conclusions:Galectin-3,Wnt pathway related protein β-catenin and Cyclin D1 are correlated with each other,which may provide new sight in the treatment of salivary gland tumor.

    • Effect of high intensity focused ultrasound on immune function of patients with hilar cholangiocarcinoma

      Abstract:Objective:To observe the effect of high intensity focused ultrasound(HIFU) on immune function of patients with hilar cholangiocarcinoma. Methods:HIFU treatment was performed on 32 patients with hilar cholangiocarcinoma. Tests were carried out to find change of T lymphocyte subsets in peripheral blood(CD3+,CD4+,CD8+),cytoactive rate of NK cells and titer of IgG,IgA and IgM before and after HIFU treatment. Results:After HIFU treatment,cytoactive rate of CD3+,CD4+、NK were obviously heightened,however,that of CD8+ was descended(P<0.05). Titer of IgG,IgA and IgM was increased obviously(P<0.01). Conclusion:HIFU posses some ef-fects in adjusting immune damage in cells,overcoming tumor immunity escaping and improving immune function in organism.

    • Clinical efficacy of minimally invasive Nuss surgery in the treatment of patients with pectus excavatum at different ages

      Abstract:Objective:To investigate the clinical efficacy of minimally invasive Nuss surgery in the treatment of patients with pectus excavatum at different ages. Methods:Twenty patients aged between 3-6(not including 6),20 patients aged between 6-12(nog in-cluding 12) and 20 patients aged older than 12 were selected and treated with Nuss procedure. Comparison was made between the three groups concerning the perioperative situation,postoperative complications and postoperative orthopedic effects. Results:There was no significant difference in operation time,amount of blood loss,postoperative hospital stay among groups of different ages. But the older the patients were,the longer time the analgesic and ambulation required,which were of significantly differences. There was no fatal postoperative complication in the three groups and there was no significant difference in complication incidence among groups of different ages. The orthopedic effects were fine except one case with age older than 12. With the growing of age,the psychological obstacles were increased,therefore,the orthopedic effects became less satisfactory. Conclusions:Nuss surgery can achieve fine ortho-pedic effects,which varies inversely with age. 3-6 years old is the ideal age for performing Nuss procedure surgery.

    • Clinical analysis of autoimmune response in patients with myocardial disease

      Abstract:Objective:To calculate the rates of anti-myocardial antibody(AMA) in the patients with myocardial disease and to ex-plore the relationship between autoimmune response and myocardial injury. Methods:Totally 192 cases of cardiac damage were en-rolled in the study,including 44 cases of viral myocarditis(VMC),among which 9 cases were severe acute myocarditis,38 cases of di-lated cardiomyopathy(DCM),40 cases of rheumatic heart disease(RHD),among which 10 cases had active rheumatism,30 cases of systemic lupus erythematosus(SLE) and 40 cases of coronary heart disease(CHD). Anti-myocardial antibodies of these serum sam-ples were detected by indirect immunofluorescence assay. Results:The positive rates of anti-myocardial antibody in VMC,DCM,RHD,SLE and CHD group were 27.3%,23.7%,17.5%,16.7% and 2.5% respectively. There was no statistical significance between patients with serious acute virus myocarditis and other types of virus myocarditis. There was no significant difference in positive anti-myocardial antibody between RHD patients with active rheumastism and those without active rheumatism. Conclusions:Myocardial diseases are correlated with autoimmune response. Detecting the antimyocardial antibody in serum of patients could not only help to know the degree of myocardial injury but also guide the clinical treatment.

      Abstract:Objective:To investigate the causes of traumatic injury induced visual impairment and to discuss the mechanism of the in-jury and the main points during the identification. Methods:The clinical data were collected from Chongqing institute of forensic medicine(2009-2012). The clinical data of 200 cases of visual impairment were analyzed concerning the factors of age,sex,causes,pathology and severity degree of the injury. Results:The ratio of male to female was 2.92∶1; the ratio of left eyes injury to right eyes injury was 1.50∶1;the age ranged within 2-83 years old with the highest incidence in 40-49 years old group. The major cause(45.0%) of the injury was traffic accident followed by blunt force injury. The traffic accident resulted in optical nerve injury and blunt force in-jury resulted in ocular contusion. Low vision disability in grade 1 and 2 accounted for 41.2% and blind level(grade 3) accounted for 47.7% in vision identification. Conclusions:Traumatic injury is the main cause of visual impairment and the incidence of blindness is high. The main mechanism is the optic never injury caused by traffic accidents. In the identification,we should notice whether the visual impairment after injury is directly caused by the trauma. The damage of cornea and secondary glaucoma are always the causes of low vision disability in grade 1,but when the complication comes,the harm gets worse obviously.

    • Clinical application of OMOM capsule endoscopy in 251 cases

      Abstract:Objective:To investigate the diagnostic value of capsule endoscopy(CE) in patients with gastrointestinal diseases and to guide the reasonable application of capsule endoscopy. Methods:The clinical data of 251 patients underwent OMOM capsule en-doscopy with telephone follow-ups were retrospectively analyzed. Results:There were significant differences in the patients with ob-scure gastrointestinal bleeding between the elderly group and the non-elderly group(45.3% vs. 26.7%,P=0.005 7). Meanwhile,there were significant differences in the patients with abdominal pain or diarrhea between the elderly group and the non-elderly group (54.7% vs. 71.1%,P=0.015 9). The rate of complete small-bowel examination was 93.2%. The gastric transit time in complete small-bowel examination was significantly shorter than that in the non-complete small-bowel examination group(47.4 min vs. 66.2 min,P=0.043 2). CE showed that the risk of non-complete small-bowel examination was bile reflux(OR=4.49). Metoclopramide(10 mg,intramuscular injection) did not exerted effect on the small bowel transit time(298.3 min vs. 313.3 min),but it can reduce the gas-tric transit time of the capsule. The positive detection rate of small bowel diseases was up to 94.9% in the patients with obscure gas-trointestinal bleeding(75/79),most commonly in vascular malformation(31.6%). The positive detection rate was 44.4%(8/18) for high risk population of small intestine ancylostomiasis;85.7%(144/168)for abdominal pain or diarrhea group with mucosal inflammation was frequent seen(26.8%). There were no significant differences in the detection rate of small bowel diseases among the elderly group,the children group and the middle-aged group(89.1% vs. 100% vs. 91.7%). The detection rate of small bowel diseases was significantly higher in the high definition capsule endoscopy group than that in the 06 capsule endoscopy group(96.7% vs. 86.4%,P=0.032 3). All patients were well tolerated and the capsule and successfully discharged. Conclusion:The manipulation of capsule endoscopy is simple,safe and effective and it is of important value in the diagnosis of small intestinal diseases. Metoclopramide (intramuscular in-jection) can improve the complete rate. Parasite elimination can be done before CE exam for high risk population of small intestine ancylostomiasis.

    • Bi-weekly versus tri-weekly chemotherapy regimens of Taxanes for advanced non-small cell lung cancer:a systematic review

      Abstract:Objective:To evaluate the effectiveness and safety of bi-weekly versus tri-weekly chemotherary regimens of Taxanes for non-small cell lung cancer(NSCLC). Methods:Relevant randomized controlled trials(RCTs) of bi-weekly versus tri-weekly chemotherapy regimens of Taxanes for NSCLC from databases of PubMed,Embase,Cochrane Library,Web of Scinece,VIP,Wanfan-data and CNKI were searched. The retrieval time was from the building time of each database to July 25,2011. The articles fit the in-clusion criteria were selected and the information in the articles was extracted. The quality of these included studies in accordance with the quality evaluation criteria recommended by the Cochrane Collaboration was evaluated and then analyzed statistically by RevMan 5.0 software. Results:Eight RCTs and 624 patients were included. Meta analysis results suggested that the bi-weekly regi-men was more effective in decreasing the incidences of leukopenia(RR=0.49,95%CI:0.38-0.62,P<0.01),nausea and vomiting(RR=0.60,95%CI:0.44-0.81,P<0.01) and alopecia(RR=0.35,95%CI:0.16-0.77,P<0.05) than tri-weekly regimen. There was no statisti-cal difference between bi-weekly regimen and tri-weekly regimen in effective rate(RR=1.09,95%CI:0.88-1.35,P=0.44)、1-year sur-vival rate(RR=1.16,95%CI:0.91-1.47,P=0.22) and liver toxicity(RR=0.26,95%CI:0.04-1.56,P=0.14). Conclusions:The effec-tive rate and 1-year survival rate are similar between the two chemotherary regimens,but toxicity incidence of bi-weekly chemother-ary regimen is lower than that of tri-weekly chemotherary regimen.

    • Efficacy and safty of Amiodarone in preventing atrial fibrillation after transthoracic esophagectomy

      Abstract:Objective:To determine the efficacy and safety of Amiodarone in preventing atrial fibrillation(AF) after transthoracic esophagectomy. Methods:Seventy-two patients undergoing transthoracic esophagectomy consecutively were randomly divided into ex-perimental group(n=36) and control group(n=36). Amiodarone-treated patients received Amiodarone in a dose of 200 mg three times per day for 7 d,then 200 mg per day until the day of surgery. The incidence of AF,hospital duration,occurrence of other complications and adverse events in the two groups were compared. Results:There was no significant difference between experimental and control groups in demographic characteristics. The incidence of AF was lower in experimental group than in control group(16.7% vs. 39.8 %,(112±22) times/min vs. (134±29) times/min,P<0.05). There was no significant difference between the groups in incidences of postoperative complications and hospital duration. Conclusion:Amiodarone prophylaxis can significantly reduce the incidence of AF after transthoracic esophagectomy.

    • Inflammatory pseudotumor of the lung:CT findings and misdiagnosis analysis

      Abstract:Objective:To study the CT features of inflammatory pseudotumor of the lung(IPL) and to improve the understanding of IPL. Methods:CT findings were retrospectively reviewed in 26 patients with IPL proved by pathology. Results:The lesions of 20 pa-tients were located at the right lung and those of 6 patients were at the left lung. The lesions usually presented different appearances:19 cases of irregular fan-shape,7 cases of spherical shape,a knife cut edge in 17 cases and 5 cases of low density halo-shape. The density of the lesions was homogeneous in 14 cases and heterogeneous in 12 cases,with CT value of 12-41 HU;4 cases were highly enhanced,with CT value reaching more than 100 HU;9 cases were significantly enhanced on the edge;6 cases were moderately en-hanced and the CT value was 70-80 HU. Pleural change:wide base of 14 cases with adjacent pleural thickening. Seven patients with primary lymph node enlargement. Conclusions:The main CT features of IPL are as follows:the lesions were usually single,adjacent pleural,fan-shaped and highly enhanced;the edge has the burr. The comprehensive analysis of clinical history and CT imaging is helpful to improve the accuracy of diagnosis of this disease,and reduces the rate of misdiagnosis

    • Clinicopathological analysis of stomach glomus tumor in 6 cases

      Abstract:Objectve:To investigate the clinicopathological characteristics,differential diagnosis and prognosis of stomach glomus tu-mors. Methods:Totally 6 cases of stomach glomus tumors were studied by histopathological observation and immunohistochemical staining. Results:The study included 5 cases of benign tumors and 1 case of potential malignant stomach glomus tumor which was hard to predict. The tumor cells were uniform-sized,round or polygonel,arranged around the blood vessels. Immunohistochemical study demonstrated that tumor cells reacted positively to Vimentin,smooth muscle antibody(SMA),laminin,actin,but negatively to CD34,CD117,neuron specific enolase(NSE),AE1/AE3,CAM5.2 and carcino-embryonic antigen(CEA). Conclusions:Stomach glo-mus tumor is rare and most of the time benign. It can be easily misdiagnosed by frozen section and clinical and cytological tests so that more attention should be paid on the disease and unnecessary surgical therapies should be reduced.
