    • Study of recombinant human parathyroid hormone to accelerate orthodontic tooth movement after anterior maxillary osteotomy

      Abstract:Objective:To study the feasibility and regulation mechanism of recombinant human parathyroid hormone(rhPTH) for pro-moting orthodontic tooth movement after the maxillary anterior osteotomy. Methods:Forty-eight rabbits were used to establish the model of anterior maxillary osteotomy and orthodontical movement of right maxillary first molars. They were randomly divided into the experimental group and the control group. The experimental group received subcutaneous injection of rhPTH 20 μg/kg,while the control group received the normal saline every other day. The mesial movement distance of the right maxillary first molars was measured on the 5th,7th,14th and 21th day. The mesio-periodontium of moving teeth were taken and tartrate resistant acid phosphatase(TRAP) staining of osteoclast count,immunohistochemistry and fluorescence quantitative polymerase chain reaction(qPCR) of sclerostin(SOST) and bone morphogenetic protein-2(BMP-2) were performed. Results:During the same period,the moving distance and speed of the first molars in the experimental group were faster than those in the control group(P<0.05). In the mesial periodontal tissue of right maxillary first molars,the osteoclast count in the experimental group was significantly larger than that in the control group(P<0.05). According to the immunohistochemistry and qPCR,the expression of SOST mRNA and protein of the mesial periodontal tissue of right maxillary first molars in the experimental group was significantly higher than that in the control group(P<0.05),while the expression of BMP-2 mRNA and protein in the experimental group was lower than that in the control group(P<0.05). Conclusion:In the proximal pressure side of the orthodontic tooth after maxillary anterior osteotomy,subcutaneous injection of rhPTH on every other day may in-crease the alveolar bone remodeling to accelerate the movement speed of orthodontic tooth by upregulating the expression of SOST,downregulating the expression of BMP-2 and increasing the re-construction of alveolar bone.

    • Intervention of S-adenosyl-L-methionine on oxidative stress in rats with autoimmune emphysema

      Abstract:Objective:To study the intervention effect of S-adenosyl-L-methionine(SAM) on oxidative stress in rats with autoimmune emphysema. Methods:Thirty SD male rats with age of ten weeks were randomly divided into three groups:the normal control group(n=10),the emphysema group(n=10) and the SAM group(n=10). Rats in the emphysema group and the SAM group were treated with intraperitoneal injection of human umbilical vein endothelial cells plus complete Freund’s adjuvant to establish autoimmune emphy-sema models,while in the normal control group were treated with intraperitoneal injection of the same dosage complete Freund’s adju-vant. In addition,rats in the SAM group were also treated with intraperitoneal injection of SAM of 15 mg/(kg·d). After 21 days,tissue sections of right lung were stained by hematoxylin eosin(HE),pathological changes were observed,and mean linear intercept(MLI) and mean alveolar number(MAN) were measured. At the same time,malondialdehyde(MDA),superoxide dismutase(SOD),activities of glutathione peroxidase(GSH-Px),and the glutathione(GSH) in serum were detected by the spectrophotometry. Levels of anti-endothelial cell antibody(AECA) in serum were detected by using ELISA. Results:MLI in the emphysema group was higher than that in the normal control group,MAN in the emphysema group was lower than that in the normal control group,while MLI and MAN in the SAM group were both in the middle when com-pared with those two groups,with statistically significant differ-ences(P=0.000;P=0.000). GSH concentration,SOD activity,and GSH-Px activity in the emphysema group were decreased than those in the normal control group(P=0.000;P=0.000;P=0.000),while in the the SAM group were in the middle when compared with those two groups(P=0.000). MDA concentration in the emphysema group was higher than that in the normal control group,while in the SAM group were in the middle when compared with those two groups(P=0.000). Conclusion:SAM may inhibit the for-mation of autoimmune emphysema in rats by increasing the production of GSH concentration,SOD activity,and GSH-Px activity,and decreasing the level of MDA.

    • Establishment of exercise induced pulmonary hypertension rat model induced by monocrotaline and early intervention effect of tadalafil

      Abstract:Objective:To establish exercise induced pulmonary hypertension(EiPH) rats model,diagnose in the early phase and treat as soon as possible,so as to assess the pulmonary hypertension(PH) progression,pulmonary artery remodeling and survival rate. Methods:Sixty male Sprague Dawely(SD) rats,with weight 200 to 220 g,were intraperitoneally injected with monocrotaline(MCT,60 mg/kg). After injecting MCT,the mean pulmonary artery pressure(mPAP) under resting and exercising condition and maximum mPAP(mPAPmax) under exercising condition on the 1st,3rd,5th,7th and 9th day were measured. Rats with resting mPAP<25 mmHg and exercising mPAPmax>30 mmHg were classified into EiPH group;rats with resting mPAP≥25 mmHg were randomly divided into resting PH(rPH) group and PH group;rats with injection of saline were classified into control group. Rats in the EiPH and the rPH group were given same dose tadalafil(T) by gavage for two weeks. Echocardiography was performed at the end of experiment,right heart catheter was improved to measure the pulmonary artery pres-sure and the cardiopulmonary tissue was taken and kept. Results:On the 7th day after MCT injection,the exercising mPAPmax in the MCT group was higher than that in the control group[(38.5 ± 2.7) mmHg vs. (27.24 ± 0.46) mmHg,P=0.000]. On the 35th day after MCT injection,resting mPAP in the rPH+T group was increased[(26.67 ± 0.11) mmHg vs. (35.23 ± 0.47) mmHg,P=0.000],right ventricular hypertrophy index was raised(0.34 ± 0.01 vs. 0.46 ± 0.02,P=0.004) and the percentage of pulmonary artery wall thickness(44.4% vs. 55.6%,P=0.008) was magnified. Conclusion:The EiPH rat model was successfully established. According to tadalafil intervention experiment,effectiveness of lowering blood pressure and inhibiting pulmonary vessel remodeling in the early phase is better than treating with a same dose drugs in the late phase.

    • Therapeutic effect of low intensity ultrasound-mediated nanobubbles loaded with double suicide gene on bladder cancer in mice

      Abstract:Objective:To prepare nanobubbles loaded with colicytocinedeaminase-thymidinekinase(CD-TK) for treating bladder cancer in mice. Methods:Nanobubbles loaded with CD-TK double suicide gene were prepared by thin layer method and cationic nanobub-bles loading CD-TK double suicide gene were synthesized. Nanobubbles’ shape was observed through light microscope and the par-ticle diameter was measured via Malvin laser particle size analyzer. The mice model with bladder cancer was established,ultrasound was used to irradiate the tumor site and the double suicide gene was transfected. Intraperitoneal injection of atoxic prodrug was con-tinuously given to each group for one week. The longest and shortest diameters of each nude mouse’s tumor were recorded,and the tumor’s volume changes were calculated and recorded. Tumor specimens were taken at the fifth week. Apoptosis and proliferation of tumor cells were evaluated by immunofluorescence. Results:The nanobubbles loading CD-TK double suicide gene were successfully prepared in this study. Ultrasound-mediated nanobubbles loaded with CD-TK can successfully transfect suicide gene,inhibiting the proliferation and apoptosis of tumors. Conclusion:Nanobubbles loaded with CD-TK may be a new therapy for bladder cancer.

    • Expression and significance of lncRNA-UCA1,HMGB1/RAGE in non-small cell lung cancer

      Abstract:Objective:To clarify the expression of long non-coding RNA-Human urothelial carcinoma associated 1(LncRNA-UCA),high mobility group box-B1(HMGB1),receptor for advanced glycation end-products(RAGE) and nuclear factor kappa-B(NF-κB) p65 in human non-small cell lung cancer(NSCLC) tissues and normal tissues,and to explore its relationship with the clinical fea-tures and pathological types of NSCLC. Methods:Forty patients with NSCLC admitted to our hospital from May 2018 to August 2018 were enrolled. Cancer tissues and adjacent normal tissues of 40 patients were respectively collected. The expression of HMGB1 and RAGE in cancer tissues and normal tissues 10 patients were detected by immunohistochemistry. The expression of HMGB1,RAGE,NF-κB p65 protein in cancer tissues and normal tissues of 12 patients was detected by Western blot. The mRNA expression of LncRNA-UCA1,HMGB1,RAGE and NF-κB p65 in cancer tissues and normal tissues of 40 patients were determined by quantitative real-time polymerase chain reaction(qRT-PCR). Results:Immunohistochemistry and Western blot analysis showed that expression of HMGB1(1459.72±366.67) and NF-κB p65(479.52±265.07) in the NSCLC tissues was significantly higher than that in the adjacent normal tissues(847.60±313.66,135.53±109.61)(t=5.470,P=0.000;t=4.063,P=0.002),while expression of RAGE(127.49±43.91) in the NSCLC tissues was significantly lower than that in the adjacent normal tissues(257.61±64.84)(t=-11.461,P=0.000). The qRT-PCR detection showed that expression of LncRNA-UCA1(4.85±2.31),HMGB1(4.64±2.10) and NF-κB p65(4.22±2.10) in the NSCLC tissues was significantly higher than that in the adjacent normal tissues(1.00±0.00),while expression of RAGE(0.41±0.19) in the NSCLC tissues was significantly lower than that in the adjacent normal tissues(1.00±0.00)(t=-19.754,P=0.000). The expression of LncRNA-UCA1,HMGB1 and NF-κB p65 in the adenocarcinoma(2.93±1.40,3.06±1.36,2.17±0.87) was higher than that in the squamous cell carcinoma(6.77±2.22,6.16±2.20,5.96±2.90)(t=3.722,P=0.001;t=3.114,P=0.004;t=3.348,P=0.002),in the cancer tissue more than 3 cm(6.70±2.14,6.28±1.80,6.07±2.17) was higher than that in tissues less than 3 cm(3.11±1.40,3.21±1.43,2.28±0.86)(t=-5.367,P=0.000;t=-5.181,P=0.000;t=-5.623,P=0.000),and in cancer tissues with lymph node metastasis(6.38±2.25,6.04±2.25,5.72±2.81) was higher than that in cancer tissues without lymph node metas-tasis(2.70±1.30,3.23±1.38,2.24±0.92)(t=2.377,P=0.023;t=2.404,P=0.021;t=2.410,P=0.021). The expression of RAGE in adenocarcinoma tissues(0.54±0.18) was lower than that in squamous cell carcinoma tissues(0.28±0.12)(t=-3.049,P=0.004),in tumor tissues more than 3 cm(0.28±0.11) was lower than that in tumor tissues less than 3 cm(0.54±0.18)(t=3.870,P=0.000),and in cancer tissues with lymph node metastasis(0.27±0.11) was lower than that in cancer tissues without lymph node metastasis(0.52±0.18)(t=-2.541,P=0.015). Pearson correlation analysis showed that in cancer tissues of patients with NSCLC,HMGB1 was positively correlated with expression of LncRNA-UCA1 and NF-κB p65(r=0.754,P=0.000;r=0.704,P=0.000),while RAGE was negatively correlated with expression of HMGB1 and NF-κB p65(r=-0.689,P=0.000;r=0.704,P=0.000). Conclusion:The high expression of LncRNA-UCA1 is related to the occurrence and development of NSCLC;LncRNA-UCA1 may play a role in influencing HMGB1/RAGE expression and NF-κB pathway.

    • Ovarian functional reconstruction in rats with premature ovarian failure through hyperbaric oxygen combined with bone marrow mesenchymal stem cell transplantation

      Abstract:Objective:To study the ovarian functional reconstruction in rats with premature ovarian failure(POF) through hyperbaric oxygen(HBO) combined with bone marrow mesenchymal stem cells(BMSCs) transplantation. Methods:Female sprague dawley(SD) rats were subjected to intraperitoneal injection of cyclopho sphamide(CTX) to induce POF. Successful modeling(36 rats) were ran-domly divided into the model group,the BMSCs transplantation group,and HBO+ BMSCs transplantation group,12 in each group. Another 12 rats were taken from the blank group. After the BMSCs transplantation group and the HBO+BMSCs transplantation group were given individual intervention,rats estrus cycle,serum E2,FSH hormone level,the amount of follicles at all levels were recorded respectively,and ovarian microstructure,distribution of transplanted cells in ovarian tissue and reproductive ability were observed so as to evaluate the ovarian function. Results:The recovery of estrus cycle in the HBO+BMSCs transplantation group was better than that in the BMSCs transplantation group. E2 in the HBO+BMSCs transplantation group(82.463±4.108) was higher than that in the BMSCs transplantation group(77.233±3.714)(P=0.047) and FSH in the HBO+BMSCs transplantation group(21.292±2.458) was lower than that in the BMSCs transplantation group(24.574±2.389)(P=0.043). Compared with the BMSCs transplantation group (13.50±1.43),(5.40±0.70),the number of GF(11.50±1.27) MF(4.50±0.53) in the HBO+BMSCs transplantation group increased at varying degrees(P=0.024,0.028). Compared with the BMSCs transplantation group(41.30±3.20,13.30±1.89),the number of PF(46.00±3.74),AF(16.10±2.18) in the HBO+BMSCs transplantation group decreased at varying degrees(P=0.044,0.040). The ex-pression of green fluorescence in the HBO+BMSCs transplantation group was more than that in transplantation group after 30 days of transplantation. SD rats in the HBO+BMSCs transplantation group had more offspring than that in the BMSCs transplantation group. Conclusion:The HBO+BMSCs transplantation has a synergistic effect on functional reconstruction of POF,and it may play a safe and effective role in the treatment of POF.

      Abstract:Objective:To explore the molecular mechanism of long-chain non-coding RNA PANDAR(LncRNA PANDAR) to promote the growth and metastasis of thyroid cancer. Methods:Firstly,real-time quantitative PCR was used to detect the expression of PANDAR in serum and cancer tissues of patients with thyroid cancer;measures like MTT,flow cytometry and Transwell were used to detect the effect of PANDAR on growth,apoptosis and migration of thyroid cancer cells. The dual luciferase reporter gene assay was used to determine that miR-637 was the target of PANDAR and to determine the role of miR-637 in PANDAR on growth,apoptosis and mi-gration of thyroid cancer cells. Results:Expression of PANDAR was significantly increased in serum and cancer tissues of patients with thyroid cancer,and PANDAR was able to significantly promote cell growth and migration,and inhibit apoptosis. The target of PANDAR was miR-637 and miR-637 was able to reverse PANDAR-induced cell proliferation,metastasis and apoptosis. Conclusion:PANDAR regulates proliferation and metastasis of thyroid cancer by targeting miR-637.

    • Investigation of the humoral immune index levels among Tibetans and Hans in Changdu,Tibet

      Abstract:Objective:To investigate the levels of the humoral immune index among Tibetans and Hans in Changdu,Tibet,and com-pare the differences between the two groups,and that between samples in Tibet and Chongqing. Furthermore,to discuss the establish-ment of local reference intervals. Methods:Serum samples were collected from 392 subjects who underwent physical examination in The Changdu People’s Hospital of Tibet from April to September 2018,including 195 Tibetans and 197 Hans. Serum IgA,IgG,IgM,complement C3 and C4 levels were detected by immune scatter turbidity method,which were compared with the humoral immune index levels of 200 subjects who underwent physical examination in The First Affiliated Hospital of Chongqing Medical University,so as to analyze the differences among different regions,nationalities and genders. Results:The levels of complement C3(Z=-11.900,P=0.000) and IgM(Z=-3.063,P=0.002) in Tibet were higher than those in Chongqing while the level of IgA was lower than that in Chongqing(Z=-4.287,P=0.000) and there were not significant differences in the levels of complement C3 and IgG. In Tibet,the C3 level of the Tibetans was higher than that of the Hans(Z=-2.799,P=0.005),and the rest showed no significant difference. The levels of IgG(t=-3.787,P=0.000) and IgM(Z=-6.736,P=0.000) in female Tibetans were significantly higher than those in the males,and the levels of IgG(Z=-3.817,P=0.000) and IgM(Z=-3.356,P=0.001) in female Hans were also higher than those in male Hans. Addi-tionally,the C3 level in male Tibetans was higher than that in male Hans(Z=-3.489,P=0.000) while the IgM level in male Tibetans was lower than that in male Hans(Z=-2.521,P=0.012). However,there were no significant differences between Tibetan female popu-lation and Han female population in humoral immune index levels. Conclusion:The humoral immune index levels of Tibetan and Han people in Changdu,Tibet are different from those in Chongqing,and differences between different nationalities and genders are also present. Therefore,specific reference intervals should be established according to the healthy population of each group.

    • Analysis of association between marital status at diagnosis and prognosis in 142 007 patients with breast cancer based on SEER database

      Abstract:Objective:To investigate the association between marital status at diagnosis and breast cancer specific survival(BCSS). Methods:Data of patients who were diagnosed with breast cancer between 2010 and 2014 were extracted from the Surveillance,Epi-demiology,and End Results(SEER) database. Association between marital status(married,single,divorced,separated and widowed) and BCSS was retrospectively analyzed by univariate and multivariate Cox proportional hazards models. Results:A total of 142 007 patients were enrolled,with a median follow-up of 28 months(IQR:2-59 months),and 6685 patients were died with specific breast cancer. There were significant imbalances in clinicopathologic characteristics[age,tumor stage,histological grade,estrogen receptor(ER),progesterone receptor(PR),human epidermal growth factor 2(HER2),molecular type,chemotherapy and radiation therapy] between patients with different marital status(all P<0.001). Multivariate Cox proportional hazards model analyses showed that the single(HR=1.26,95%CI=1.18 to 1.35,P<0.001),the divorced(HR=1.11,95%CI=1.07 to 1.16,P<0.001),the widowed(HR=1.10,95%CI=1.03 to 1.18,P<0.001) and the separated(HR=1.18,95%CI=1.16 to 1.20,P<0.001) were bad prognostic factors for breast cancer patients when compared with married patients. Subgroup analysis showed that separation had no influences on patients with a subtype of HR-/HER2+,HR+/HER2+ or HR-/HER2-,and divorce was not associated with the BCSS in HR-/HER2- subtype patients(P>0.05). After the correction of age,single was a poor prognostic factor for breast cancer patients of all ages. Divorce was a poor prognostic factor for patients aged 60 to 69 years old(HR=1.18,95%CI=1.10 to 1.27,P<0.001). Separation influenced the prognosis of patients aged less than 49 years old and 60 to 69 years old(P<0.05). Widowhood was a poor prognostic factor for patients aged 70 years old or older(HR=1.10,95%CI=1.07 to 1.13,P<0.001). Conclusion:Marital status at diagnosis is significantly correlated with prognosis of breast cancer and has different influences on the prognosis of patients with different age groups and molecular subtypes.

    • Clinical and prognostic analysis of 199 children with acute interstitial nephritis

      Abstract:Objective:To analyze the clinical feature and treatment of acute interstitial nephritis(AIN),so as to enhance the knowledge of AIN. Methods:Clinical data,disease cause,clinical features,laboratory and nephritic pathological results,and treating outcome of 199 patients with AIN in our hospital from January,2010 to December,2017 were analyzed. Results:Among 199 patients,180 pa-tients were infection-induced AIN,with the most common of respiratory infections,following by digestive infection; with main etiology of virus infection,including EB virus,Coxsackie virus,respiratory syncytial virus and so on. And 125 patients were caused by drugs (all were post-infection administration),with the most common drugs of anti-inflammatory drugs(NSAIDS) and antibiotics. Almost all children had renal involvement,including edema,oliguria,hematuria,proteinuria and aseptic leukocyturia to different degrees. Besides,69 patients had acute renal insufficiency. Four patients underwent renal biopsy and results showed that the majority was tubulointerstitial lesions and the glomerulus and renal vessels were basically normal. All patients were treated with removal cause and symptomatic treatment after admission. A total of 46 patients were treated with blood purification because of acute renal failure(ARF) or poisoning and 6 of them were treated with combination of glucocorticoid,after which renal function and urine tests returned to nor-mal in the short term. Conclusion:The main cause inducing AIN in children is infection,among which viral infection takes the domi-nant position,followed by drugs. The main clinical manifestations is unexplained renal function decline and abnormal urine test,among which severe patients may have AFR. Timely diagnosis and cause remove are key to treatment. Blood purification and short-term glucocorticoid therapy are needed when necessary and most of them have a good prognosis.

    • Effects of high-flow nasal cannula for acute hypoxaemic respiratory failure patients after thoracotomy

      Abstract:Objective:To investigate the therapeutic effect of high-flow nasal cannula(HFNC) oxygen therapy after thoracotomy in patients with acute hypoxaemic respiratory failure(AHRF). Methods:A total of 53 patients who received thoracotomy in our hospital from January 1,2016 to December 31,2017 and had AHRF after discharging from ICU were retrospectively analyzed and divided into the general oxygen therapy group(n=31) and HFNC group(n=22). Saturation of peripheral oxygen(SpO2),heart rate(HR) and respiration rate(RR) both before and after treatment were recorded at 2,12,48 h;indicators such as pharyngalgia/rhinodynia incidence,re-intubation rate,hospitalization time,in-hospital mortality and sputum drainage amount were also recorded. Results:When compared with the general group,post-therapy SpO2 and sputum drainage amount in HFNC group was significantly improved;heart rate,respiratory rate,reintubation rate and pharyngalgia/rhinodynia incidence were significantly decreased in HFNC group(P<0.05). Conclusion:For treating AHRF,HFNC has a better therapeutic effect than the general oxygen therapy,which can significantly improve the oxygenation in AHRF patients after thoracotomy,increase sputum drainage amount and reduce re-intubation rate;after HFNC,patients’ clinical symptoms are significantly improve and patients have a good tolerance. Therefore,HFNC deserves application in the general ward.

    • Correlation among expression level of circulating miR-126,risk of acute respiratory distress syndrome,and septic severity and prognosis in patients with sepsis

      Abstract:Objective:To assess the predictive role of plasma microRNA(miR)-126 on risk of acute respiratory distress syndrome (ARDS) in patients with sepsis,and to investigate the correlation of miR-126 and sepsis severity,inflammation level and prognosis. Methods:A total of 172 patients with sepsis were continuously enrolled. Peripheral blood samples of patients were collected within 24 hours after admission,and plasma was separated;expression level of miR-126 was detected by quantitive polymerase chain reaction. Expression levels of inflammatory cytokines[including tumor necrosis factor-α(TNF-α),interleukin(IL)-1β,IL-6 and IL-17) in plasma were detected by enzyme-linked immune sorbent assay(ELISA). Meanwhile,the 28-day mortality was recorded as well. Results:Among them,50 patients(30.8%) had ARDS during hospitalization,and plasma miR-126 expression was significantly elevated in ARDS patients when compared with non-ARDS patients(P<0.001). According to the receiver operating characteristic curve(ROC) analysis,expression level of plasma miR-126 was able to differentiate ARDS patients form non-ARDS patients,with area under the ROC(AUC) of 0.741(95%CI=0.666-0.815). In accordance with the multivariate logistic regression model analysis,high expression of miR-126(P<0.001) was the independent predictive factor for relative-high onset-risk of ARDS. In addition,of all sepsis patients,plasma miR-126 expression was positively correlated with levels of serum creatinine(P=0.004),C reactive protein(P<0.001),procal-citonin(P<0.001),acute physiology and chronic health evaluation Ⅱ(APACHE Ⅱ) score(P<0.001),sequential organ failure as-sessment(SOFA) score(P<0.001),and inflammatory factors of TNF-α(P=0.001),IL-1β(P=0.002),IL-6(P=0.011) and IL-17(P=0.002). Moreover,the expression level of plasma miR-126 in the dead was significantly higher than that in the survivors(P<0.001). According to the ROC curve analysis,it was able to predict that patients with sepsis had a high 28-day mortality risk,with AUC of 0.686(95%CI=0.601-0.771). Conclusion:Higher circulating miR-126 expression can predict the higher ARDS in-cidence in patients with sepsis,and can serve as a biomarker for disease surveillance and patients’ prognosis in sepsis.

    • A study of the relationship between etiology and clinical features of children with severe hand,foot,and mouth disease

      Abstract:Objective:To analyze the differences in clinical features of severe hand,foot,and mouth disease caused by different en-teroviruses(EV),and to provide scientific evidence for its clinical diagnosis,treatment,and prevention. Methods:A total of 421 children admitted and diagnosed with severe hand,foot,and mouth disease in Children’s Hospital of Chongqing Medical University from April 2016 to April 2017 were selected,and quantitative real-time PCR was used to detect EV. Excluding 13 children with unclear re-sults,408 children were included in the final study. According to EV strains,the children were divided into EV71 group,non-EV71 group,and EV-negative group to compare their clinical features. Results:Among the 408 children,137(33.59%) were positive for EV71,212(51.96%) were positive for non-EV71,and 59(14.46%) were negative for EV. The EV71 group had a significantly longer duration of fever than the non-EV71 group(P=0.000) and the EV-negative group(P=0.017),and had a significantly higher inci-dence of limb tremor(P=0.002) and headache(P=0.003) than the non-EV71 group. The non-EV71 group had a significantly higher incidence of skin rash on the mouth(P=0.003),limbs(P=0.001),and trunk(P=0.006) than the EV71 group,but had a significantly shorter duration of skin rash than the EV71 group(P=0.000) and EV negative group(P=0.015). Conclusion:Patients with severe hand,foot,and mouth disease caused by EV71 have more severe symptoms of fever and neurological involvement than other groups,while those caused by non-EV71 have a wider distribution but a shorter duration of rashes.

    • Identifying the genetic etiology of a child with Bainbridge-Ropers syndrome

      Abstract:Objective:To analyze the genetic etiology of a child affected by severely developmental delay,atrial septal defect and a series of complicated phenotypes. Methods:The child has undergone history taking and physical examination. Genome DNA was extracted from peripheral blood samples of the proband and his parents. Candidate genes were captured with Agilent SureSelect and sequenced on an lllumina platform. Suspected mutation was identified and verified by Sanger sequencing. Results:By whole exome sequencing,we found a heterozygous frameshift mutation in proband’s ASXL3 gene(ASXL3;c.1349dupT;p.I450fs) and identified it as pathogenic variant according to the ACMG standards and guidelines for the interpretation of sequence variants. And our candidate has cartilaginous ossification of larynx and dorsiflexion of finger,which are never been reported in other Bainbridge-Ropers syndrome(BRS) patients. Meanwhile,we found a reported mital valve prolapsed 2(MVP2) pathogenic variant DCHS1;c.G7538A;p.R2513H. The function of this variant has been proved by cell and animal experiments. The knock-out of DCHS1 in zebra fish resulted in a cardiac atrioventricular canal defect and the atrial septal defect in our proband could also be attributed to DCHS1 mutation. The proband’s mutant is inherited from his father,however no cardiac function and structure abnormality were observed in his father by ultrasonic cardiogram and electrocardiogram. This might be due to nonpenetrance. Conclusion:Our study broadened the mutant and phenotype spectrum of ASXL3. And we reported the first Bainbridge-Ropers patient with other pathogenic variant.

      Abstract:Nontypeable Haemophilus influenzae(NTHi) is a conditioned pathogen colonizing the human nasopharynx. It often causes secondary infections,such as bronchitis,sinusitis,otitis media in children,and acute exacerbations of chronic obstructive pulmonary disease in adults. It has been found that a variety of immunogenic and antigenic proteins are expressed on the outer membrane of the cell wall of NTHi,which are essential for its colonization,invasion,and survival in the host. In addition,some proteins are also associ-ated with the drug resistance and host immune clearance mechanism of the bacterium. Therefore,the study of outer membrane proteins is beneficial to the development of effective vaccines and anti-infective drugs to prevent and treat infections caused by NTHi. In this paper,several outer membrane proteins are reviewed in terms of their structure,function and application.

      Abstract:To promote the exploration and research about platelet membrane-based nanomedicine delivery system,words including “platelet”,“platelet membrane”,“nanoparticle”,“drug delivery” were selected as key words;literatures related to platelet membrane-based nanomedicine delivery system from 1961 to 2019 were searched in Chinese and foreign databases,such as PubMed,CNKI and others;its characteristics,functions,development,extraction methods,drug-loading methods,clinical applications,advantages and dis-advantages were reviewed. There are many kinds of membrane proteins on platelet membranes,which interact with different types of cells and play specific physiological and pathological functions in the body. Platelet membranes were extracted by repeating freezing-thawing method,and were used to load nanoparticles through covalent conjugation,direct encapsulation or coating. At present,platelet membrane coated nanoparticles have been used in anti-tumor,anti-infection,treatments of thrombocytopenia,immunotherapy,phototherapy and so on,with a broad application prospect. Platelet membrane-coated nanomedicine delivery system has many advan-tages,such as long retention time in blood circulation,high targeting ability and good biocompatibility;at the same time,it has short-comings of complex preparation process,biosafety and stability.

      Abstract:With the aggravation of population aging,Alzheimer’s disease(AD),as an irreversible neurodegenerative disease,plagues people all over the world and brings a heavy burden to families and the society. Acupuncture plays an increasingly important role in the clinical treatment of AD with its advantages of safety,simplicity and good effect. This paper summarizes the molecular biology researches of acupuncture in the treatment of AD in the past five years and also concludes the treatment effects of acupuncture on AD achieved by regulating the level of abnormal proteins,regulating the central cholinergic system,protecting neurons,regulating energy metabolism,inhibiting inflammatory response,and increasing the level of autophagic activity. That provides a theoretical basis for the clinical application of acupuncture in the treatment of AD.

A number of studies have shown that ND involves extensive degenerative changes in the brain. Long-term exercise can im-prove corticocerebral functional activeness in aged people,which may have a potential physiological mechanism of promoting the ex-pression of brain-derived neurotrophic factor in hippocampus region and further improve the cognitive function. This paper reviewed that effects of exercise on the function of different brain regions as well as its neurological mechanism,and improvement of effective exercise and loading on cognitive function.

However,urine volume and creatinine can be relatively delayed and can be influenced by many factors. Whether the kidney of AKI/AKD patients recover or not may be the content that needs continu-ous evaluation. This article reviewed the current situation and progress of renal function renaturability from aspects of routine indicators,biomarkers and imaging methods in AKI/AKD patients.

      Abstract:Aqueous humor is produced by the ciliary body. It is an important nutrient and circulating substance in the eye. It has the function of maintaining intraocular pressure. In recent years,many scholars have detected changes in cytokine levels in aqueous humor in eye diseases such as glaucoma,uveitis,and age-related macular degeneration. Glaucoma is the first irreversible blinding disease worldwide,but the specific mechanism of its occurrence is not clear. Some researchers have suggested that changes in cytokines in aqueous humor may play an important role in the occurrence and development of glaucoma. This article reviews and analyzes the re-cent relevant literature,and it reviews the changes of several common cytokines in the aqueous humor of glaucomatous patients and their role in glaucoma.

      Abstract:Nontraumatic osteonecrosis of femoral head(NONFH) is characterized with disruption of blood supply to bone tissue and osteocyte necrosis,which is caused by a variety of risk factors. Due to its nontypical early symptoms and difficulties in diagnosis,pa-tients suffer hip joint pain and limited mobility in the late stage caused by femoral head collapse and the hip replacement will be the only choice in the final stage. Therefore,to seek for early diagnosis and treatment of NONFH has significance in early diagnosis and treatment. MicroRNA(miRNA) is a non-coding single-stranded RNA molecule that can involve in post-transcription regulation. With more advanced research techniques of miRNA and more studies on NONFH,more miRNAs are found that abnormally expressed in NONFH. This article summarized the differentially expressed miRNAs in different tissues and cells of NONFH,which has been re-ported by several studies and has consistency with those studies,and reviewed functions of these miRNAs and corresponding mecha-nisms for participating in pathogenesis.

    • Application of fast track surgery in perioperative nursing of laryngeal carcinoma

      Abstract:Objective:To explore the application effect of fast track surgery(FTS) in perioperative nursing of laryngeal carcinoma. Methods:This study recruited 77 patients with laryngeal carcinoma admitted to our hospital in July 2016 and December 2019. With the consent of the patients,the patients were randomized into control group(38 cases) and observation group(39 cases). The control group were given routine nursing,the observation group were treated with FTS perioperative nursing,and the nursing indexes of pa-tients such as postoperative complications,nutritional indicators,average postoperative length of stay,average hospitalization expens-es,patient satisfaction,scoring results of self-rating anxiety scale(SAS) and self-rating depression scale(SDS) and so on during hos-pitalization between the two groups were compared. Results:The total incidence of postoperative complications in the observation group was 5.12%,significantly lower than that(14.91%) in the control group(χ2=10.970,P=0.001). The values of hemoglobin and serum protein in the observation group before discharge were significantly higher than those in the control group(P<0.05). The average post-operative hospital stay in the observation group was(10.00±1.05) days and the average hospital stay expense was (36 943.00±2 101.90) yuan,both significantly lower than those in the control group(P<0.05). The satisfaction scores of patients in the observa-tion group were significantly better than those in the control group(P<0.05). SAS and SDS scores of patients in the observation group were significantly lower than those in the control group before and after operation(P<0.05). Conclusion:Patients with laryngeal car-cinoma treated with perioperative nursing by FTS can reduce the incidence of stress reaction and postoperative complications. The perioperative nursing of patients could acquire fast physical and mental recovery after the surgery and short-term hospital stay,which is a safe and efficient new nursing mode that is worthy of clini-cal nursing promotion and application.

    • Analysis of the effects of predictive nursing in hospitalization of patients with COVID-19

      Abstract:Objective:To explore the clinical application effects of predictive nursing in nursing management of patients with new type of coronavirus pneumonia during hospitalization. Methods:A retrospective study was used to collect the demographic and clinical characteristics of the patients. The medical team of our hospital supporting the First Hospital of Wuhan City implemented predictive nursing interventions on 83 confirmed patients with new type of coronavirus pneumonia,2 of whom were mild,73 were severe,and 8 were critical ill,admitted from February 14 to March 17,2020. Results:As of March 17,74 patients have been discharged,12 cases converted from severe to mild,2 cases were transferred to other department,3 cases were transferred to other hospital,3 cases died. One patient died of stroke at admission. The medical staffs in the clinical treatment data of 83 patients(using logistic ordered regres-sion analysis) found that: age may be the factor that promotes the development of the new type of coronavirus pneumonia(B=-0.09,P<0.05),for a total of 8 critically ill patients with an average age of>73 years;patients with a history of coronary heart disease may be at a greater risk during treatment(B=-2.39,P<0.05);smoking history may adverse the recovery progression(B=1.52,P<0.05);the abnor-mal albumin(ALB) and hemoglobin(HGB) indicators may speed up the disease process(B=-0.40,-0.05;P<0.00,0.00). Conclusion:Follow the guidelines for clinical nursing of new type of coronavirus pneumonia,foreseeably formulate timely treatment and nursing intervention protocols,propose and strictly implement medical care measures for the new type of coronavirus pneumonia have obvious effects in clinical treatment of severe and critically ill patients. The positive effects deserve clinical attention.
